Objective:Experimental study of the guiding theory of TCM etiology,simulation of Yunnan spring dryness environment factors and applied to the experimental mice,follow the test with a Decoction of the law,drug intervention treatment dry classical Sang Xing Decoction,Shashen Maidong Decoction and Sang Xing Decoction and Shashen Maidong Decoction combination.On the basis of this,according to the clinical characteristics of " domination of dryness bringing about deficiency of fluid”,“dry evil and hurting human and lung,",the general signs of experimental mice were observed,and the changes in trachea and lung tissue morphology were observed and the changes of IgG,sIgA,SP-A and SP-D contents in airway immune indexes of mice were detected.From three levels of function,structure and molecule,the effect of spring dryness environment in Yunnan on mice was preliminarily discussed,and the method and objective index were provided for the development of the animal model of Yunnan spring dryness syndrome.Method:The experimental mice were divided into 6 groups,Normal group,Model group,Negative group,Sang Xing Decoction group,Shashen Maidong Decoction group,Combination group(i.e.Sang Xing Decoction and Shashen Maidong Decoction combination group).Normal temperature and humidity in normal mice were routinely intervened and placed in an artificial climate box,and normal diet was fed.Model group,Negative group,Sang Xing Decoction group,Shashen Maidong Decoction group,and Combination group,5 groups were treated with spring dryness climateenvironment and placed in an artificial climate box,fed with fragrant dry feed.Each group was fed with different liquids each day.Among Negative group is filled with white water daily.Sang Xing Decoction did intragastric administration daily in Sang Xing Decoction group.Shashen Maidong Decoction group in daily intragastric administration with medicine Shashen Maidong decoction.Combination group in daily intragastric administration combined with Sang Xing Decoction and Shashen Maidon Decoction combinationg.The general signs of mice were observed daily,and the changes of IgG,sIgA,SP-A and SP-D in the trachea and lung tissue of mice were observed on the fifteenth day of animal tissue sampling.Result:1.Changes in general physical signs of experimental miceIn the Normal group,the mental state was good,the urine quality was clear,the quality of the stool was normal,the hair color and luster were good.Model group mice compared with Normal group,self imposed intervention factors,gradually appeared,common mental irritability,fight,tail see bloodstain scratches.Weight loss,food intake and water consumption fluctuated greatly.At the same time,Dry nails,yellow urine,stool dry and hard hair withered,a small amount of shedding.Compared with the Model group,the other 4 groups of mice showed:(1)The mice in the Negative group,Spirit is still possible,has climbing and covering phenomenon,occasionally fighting.The amount of food is roughly equal,the water volume fluctuated greatly,the urine was yellow,the texture of the stool was mild,a little dry,and the coat was still dry,and no shedding.(2)The mice of Sang Xing Decoction,the early stage saw fighting,the later stage was not seen,the body weight increased,the diet and drinking water were stable,the urine was only slightly yellow,the stool was slightly dry,and the hair was only slightly dry.(3)The mice of Shashen Maidong Decoction group were saw fight a little.Their weight increased,the urine was slightly yellow,the stool was normal,and hair was a little dry.(4)The mice of the Combination group had no fighting,weight increase,diet increase,the clear urine color,the soft stool,and the hair only slightly dry.2.Changes of trachea and lung tissue in experimental miceIn Normal group,trachea was regular in morphology,clear in lung and clear in alveolar wall.Compared with the Normal group,the mice in the Model group,some of the epithelial cells in the tracheal mucosa layer were more severe hyperplasia.Some of the lesions were damaged,shedding,thinning,missing,incomplete rupture of the mucous membrane,partial ciliated damage accompanied by shedding,and inflammatory cell infiltration in the submucosa.Thickening of bronchial wall,thickening of alveolar wall,expansion of capillaries,partial alveolar wall,interstitial congestion,blood stasis,has red blood cells,inflammatory cells and pulmonary bullae.Compared with the Model group,the other 4 groups of mice showed:(1)Negative group mice were slightly proliferated in the epithelium of the trachea mucous membrane,the cilia was shedding,and the inflammatory cells were seen occasionally,the lungs were still dilated,the alveolar wall thickened,and there were inflammatory cells and only a few pulmonary bullae.(2)Sang Xing Decoction mice had only a small amount of cilia and only some bronchi epithelium in the lungs.(3)Shashen Maidong Decoction group mice,trachea mucous layer cilium fall off,submucosa occasionally see inflammatory cells,lung bronchial epithelium mild hyperplasia with shedding,alveolar wall fracture,see a small amount of red cells and inflammatory cells.(4)Combination group mice,the trachea mucous layer part of mild hyperplasia,the submucosa still visible inflammatory cells,lungs only see a little bit A few bronchial wall ruptured,and a few red blood cells were seen.3.Changes in the immune index of airway in experimental mice3.1The content of IgG in the airway of mice: Compared with the Normal group,the IgG content in the airway of the mice in the Model group increased,but the difference was not statistically significant(P>0.05).3.2 The content of sIgA in the airway of mice: Compared with the Normal group,the sIgA content in the airway of the mice in the Model group increased significantly(P<0.01).Compared with the Model group,the sIgA content in the airway of the mice in the Negative group and the Sang Xing Decoction group all extremely significantly decreased(P<0.001);And the sIgA content in the airway of the mice in the Shashen Maidong Decoction group and the Combination group decreased,and difference was statistically significant(P<0.05).3.3The content of SP-A in the airway of mice: Compared with the Normal group,the SP-A content in the airway of the mice in the Model group increased significantly(P<0.01).Compared with the Model group,the SP-A content in the airway of the mice was decreased in both the Negative group and the Combination group,but the difference was not statistically significant(P>0.05);The SP-A content in the airway of the mice in the Sang Xing Decoction group was decreased,and the difference was statistically significant(P<0.05);The content of SP-A in the airway was increased in the mice of the Shashen Maidong Decoction group,and the difference was statistically significant(P<0.05).3.4The content of SP-D in the airway of mice: Compared with the Normal group,the SP-D content in the airway of the mice increased significantly in Model group(P<0.01).Compared with the Model group,the SP-D content in the airway of the mice in Negative group,the Shashen Maidong Decoction group and the Combination group decreased,but the difference was not statistically significant(P>0.05);And the SP-D content in the airway of the mice in Sang Xing Decoction group decreased,and the difference was statistically significant(P<0.05).Conclusion:1.In the environment of spring dryness in Yunnan,the general signs,trachea and lung tissue of mice changed correspondingly,and the contents of sIgA,SP-A and SP-D in the airway of mice were also rising.It shows that in the environment of spring dryness in Yunna,wind and dryness invade mice,affecting the physiological function of mice,and showing a series of dry symptoms.At the same time,the microstructure of mice's lungs and trachea also damaged to some extent,which affected the airway immune function of mice.2.After intervention of dryness decoction,the signs,trachea and lung tissue lesions of each group were improved to varying degrees.The sIgA,SP-A and SP-D of the airway fluid were also improved.According to the correspondence principle of Fang Zheng,we can preliminarily estimate that in this experiment,all the animals except the normal group basically conform to the animal models of dryness syndrome,that is,the Yunnan spring dryness syndrome model.The general signs,airway histopathology and airway fluid sIgA,SP-A and SP-D contents in mice can be used as an objective index for the model.3.Sang Xing Decoction group animal in many areas was better than that of Shashen Maidong Decoction and Combination group.It is suggested that the external dryness of Yunnan spring dryness syndrome is obvious.This is consistent with the results of Yunnan's spring dryness syndrome,which is "outside dryness and internal dryness coexist,and external dryness is more than internal dryness".
