Objective: This experiment simulates haze climate environment,adopts method of inhaling toxic fog to establish animal models of cold-dryness syndrome and warm-dryness syndrome of haze-induced lung injury,selects traditional Chinese medicine classical prescriptions Xingsu Powder and Sangxing Decoction to intervene the two animal models respectively,and detects the effect of traditional Chinese medicine on IL-1 ? in serum and RAGE/S100A9/NF-?B expression in lung tissue of Wistar model rats induced by haze,so as to provide objective basis for exploring effective prescriptions of traditional Chinese medicine for preventing and treating lung injury induced by haze.Methods: 40 SPF Wistar male rats were randomly divided into blank control group,warm-dryness model group,cold-dryness model group,Sangxing Decoction treatment group and Xingsu Powder treatment group,with 8 rats in each group.The blank control group was normally fed,the warm-dryness model group and Sangxing Decoction treatment group were placed in a self-made warm and dry environmentexposuring to haze,and the cold-dryness model group and Xingsu Powder treatment group were placed in a self-made cold and dry environment exposuring to haze.Each group was poisoned for 4 hours a day.Sangxing Decoction treatment group and Xingsu Powder treatment group were given the intervention of Sangxing Decoction and Xingsu Powder respectively.Behavioral changes of rats were observed during the experiment.After 4 weeks,HE staining was used to observe pathological sections of lung tissue of rats in each group.The content of IL-1? in serum of rats in 5 groups was detected by ELISA method.The content of RAGE,S100A9 and NF-?B in lung tissues of rats in each group were detected by PT-PCR,IHC and WB detection methods respectively.Results:1.Behavioral results of ratsCompared with the blank control group,the rats in the warm-dryness model group and the cold-dryness model group showd dry stool,hoarseness,withered and coarse hair.The rats in the warm-dryness model group also showed restlessness and other behaviors,while the rats in the cold-dryness model group showed laziness and other behaviors.The above behaviors of the rats in the Sangxing Decoction treatment group and Xingsu Powder treatment group were relatively light.2.HE staining results of lung tissue sections of ratsThe lung tissue morphology and structure of blank control group rats were basically normal.In the warm-dryness model group,the lung tissue structure was destroyed,fibrosis occurred,alveolar septum widened,and inflammatory cells were accumulated in the pulmonary interstitium.In the cold-dryness model group,thealveoli in some areas of the lung tissue of the rats were expanded,the alveoli septum was fractured,and the expanded alveoli fused into a larger sac.The lung tissue morphology of the rats in the Sangxing Decoction treatment group is basically normal,the alveolar space is slightly widened,and inflammatory cells are found in the interstitium.The lung tissue of rats in Xingsu Powder treatment group also had a small amount of inflammatory cell infiltration and cytoplasm exudation in alveoli.3.Results of IL-1? Content in Serum of Rats in Each GroupThe content of IL-1? in the serum of rats in the warm-dryness model group and the cold-dryness model group is higher than that in the blank control group(P<0.05).the content of IL-1? in the serum of rats in the Sangxing Decoction treatment group is significant lower than that in the warm-dryness model group(P<0.01).the content of IL-1? in the serum of rats in the Xingsu Powder treatment group is significant lower than that in the cold-dryness model group(P<0.01).4.Results of RAGE content in lung tissue of rats in each groupThe expression of RAGE mRNA in lung tissue of rats in warm-dryness model group and cold-dryness model group was significantly higher than that of blank control group(P<0.05).The expression of RAGE mRNA in lung tissue of rats in Sangxing Decoction treatment group was significantly lower than that in warm-dryness model group,(P<0.05).The expression of RAGE mRNA in lung tissue of rats in Xingsu Powder treatment group was significantly lower than that in cold-dryness model group(P<0.05).5.Results of NF-?B content in lung tissue of rats in each groupThe expression of NF-?B in lung tissue of rats in warm-drynessmodel group and cold-dryness model group was significantly higher than that of blank control group(P<0.05).The expression of NF-?B in lung tissue of rats in Sangxing Decoction treatment group was significantly lower than that in warm-dryness model group(P<0.05).Compared with the cold-dryness model group,the relative expression of NF-? B in Xingsu Powder treatment group was significantly decreased(P<0.05).6.Results of S100A9 content in lung tissue of rats in each groupThe positive expression rate of S100A9 in lung tissue of rats in warm-dryness model group and cold-dryness model group is higher than that of blank control group(P<0.05).the positive expression rate of S100A9 in lung tissue of rats in Sangxing Decoction treatment group is lower than that in warm-dryness model group(P<0.05).the positive expression rate of S100A9 in lung tissue of rats in Xingsu Powder treatment group is lower than that in cold-dryness model group(P<0.05).Conclusion:1.Animal models of warm-dryness syndrome and cold-dryness syndrome caused by haze can be established through temperature and humidity regulation and simulated haze climate environment.2.Classic prescriptions Xingsu Powder and Sangxing Decoction have certain preventive and therapeutic effects on lung injury caused by haze.
