Objective:The changes of the mouse characterization,lung organ index,trachea and lung tissue morphology,as well as airway MUC5 ac and the cilia Protein DPCD in the spring-dry environment in Yunnan were observed,and analysis of its relationship with "drying less water" and "failure of external defense " in traditional Chinese medicine.Discuss the effects of spring drying environment in Yunnan on mice from macro and micro perspectives.To a certain extent,Proving the accuracy and reliability of Yunnan spring drought animal model,and to Provide a new model and evaluation index for the further research of Yunnan spring drought experiment.Method:Thirty male Kunming mice were randomly divided into normal group,model group and Sang Xing Decoction group,with 10 mice in each.Feeding and Modeling in Animal Laboratory of Yunnan University of Traditional Chinese Medicine.Mice in the normal group were given normal climatic environment and normal diet;According to the actual climatic conditions in Kunming,the mice in the model group and the mice in the Sang Xing Decoction group were set up with the intervention of the spring dryness environment and fed with the special spicy dryness diet;Mice in the sang Decoction tang group were given Sang Xing Decoction gavage at 9 o 'clock every morning since the first day when the factors were aplied.Mice in the normal group and model group were not given gavage.Daily observation records mice body weight,food,drinking water and the status of the activity,feeding to 14 days,when the mice showed dry symptoms such as haggard and bifurcated coat,thin fingernail and nail,yellow urine color,and obvious marks of fighting and biting,carry out the experiment on the 15 th day.Observation group in mice trachea,lung tissue morphology and adhesion protein MUC5 ac,cilia DPCD protein expression in change.Results:1.Characterization Changes in MiceMice in the normal group were in good mental state during feeding,and few fight and bite marks were found.Nail ruddy color and bright,smooth fur,no withered fork,clear and transparent urine.Compared with the normal group,mice in the model group were depressed since the intervention factors were aplied,with clear fight and bite marks and increasingly obvious tail blood marks.Claw Purplish red dull thin,dry fur,bifurcation,urine color yellow.Compared with the model group,the mental state of the mice in the Sang Xing Decoction group was acceptable,and a small number of scratches apeared in the tail.Nail red lack luster,fur color Pure,occasionally bifurcated,generally yellow urine.Mice in the normal group had normal diet and drinking water,and the fluctuation was stable.Compared with the normal group,the mice in the model group ate less food(P<0.05)and drank more water(P<0.01).Although there were changes in diet and water intake in the mice of Sang Xing Decoction group,the differences were not statistically significant(P>0.05).Compared with the model group,the amount of diet increased(P<0.05)and the amount of water decreased(P<0.05)in the Sang Xing Decoction group.2.The Changes of Lung Organ Index,Trachea and Lung Tissue Morphology in MiceCompared with the normal group,the lung and organ index of the model group was significantly lower(P<0.01),and that of the Sang Xing Decoction group was lower than that of the normal group,but not statistically significant(P>0.05).Compared with the model group,the lung organ index of the Sang Xing Decoction grouP was lower(P<0.05).In the normal group,the endotracheal lumen morphology was regular,the mucosal surface was intact,and the cilia were orderly arranged.Lung tissue structure is clear,alveolar wall integrity,no hyperplasia,no edema,hyperemia Phenomenon.Mice in the model group showed local damage to the tracheal mucosa under the microscope,obvious loss and loss of cilia,occasional inflammatory cells in the submucosa,and slight expansion and hyperemia in the blood vessel wall.The bronchial wall of the lung tissue was thickened,the bronchial mucosa cilia were not aligned,the capillaries were slightly dilated,and the interstitium of the alveolar wall showed obvious congestion and congestion.The endotracheal lumen morphology of the mice in the Sang Xing Decoction group was more regular,the mucosal layer was more complete,and the cilia were orderly arranged with occasional loss.Lung tissue showed clear airway structure,regular bronchial morphology,slight thickening of capillary wall and slight hyperemia in the interstitium of alveolar wall.3.Changes of MUC5 ac Content in Airway Mucin of Experimental MiceCompared with the normal group,the MUC5 ac content of mice in the model group was significantly reduced(P<0.01).Although MUC5 ac content of mice in Sang Xing Decoction group changed,the difference was not statistically significant(P>0.05).Compared with the model group,the MUC5 ac content of mice in the Sang Xing Decoction group was significantly reduced(P>0.05).4.Changes in DPCD Content of Airway Cilia Protein in MiceCompared with the normal group,the Protein expression level of mice in the model group was significantly decreased,and the DPCD content of mice in the Sang Xing Decoction group was less than that in the normal group,but significantly increased compared with the model group.The content of DPCD in airway cilia Protein of mice from high to low was respectively normal group > Sang Xing Decoction group > model group.Conclusions:1.Under the intervention of spring drying environment in Yunnan,a series of drying symptoms apeared in mice.The mice's diet decreased and their drinking water increased significantly.At the same time,the structure of trachea and lungs of mice was damaged to a certain extent.So Yunnan spring drying environment has certain influence on the characterization of mice.2.With the intervention of Sang Xing Decoction,the structure of trachea and lung in mice was improved to some extent.According to the principle of "testing evidence by prescription",the animal model of Yunnan Spring Dryness Syndrome has certain accuracy and reliability.3.The expression levels of mucin MUC5 ac and cilia Protein DPCD in the airway of mice were significantly decreased under the intervention of spring drying in Yunnan Province.After the intervention of Sang Xing Decoction,the expression levels were increased,and the difference was statistically significant.Yunnan Spring Dry Environment has certain effects on the "cilia-mucus blanket" defense barrier of airway in mice,MUC5 ac and DPCD can be considered as evaluation indexes of animal model of spring dryness syndrome in Yunnan.
