Objective: establish a Successful hot asthma mice model;From the perspective of immune mechanism of asthma,by the way of molecular biological and molecular immunology,view the affection of Maxingxianxiongkeli on the level of IL-12 and IL-13 in serum,find the possible found targets of Maxingxianxiongkeli.Combined with the use of pathology,observe the pathological changes of lung tissue of asthmatic mice and the effect of drugs of the groups,so as to confirm the effectiveness of Maxingxianxiongkeli,and providing experimental theoretical basis for the treatment of asthma in children.Methods: Healthy,male KM laboratory mice 80,SPF,rat ages 3-4 weeks,weight(30±4)g.Randomly divided into 5 groups: blank group(group A),model group(group B),Maxingxianxiongkeli group(group C),dexamethasone group(group D),Maxingxianxiongkeli + dexamethasone group(group E),16 for each group.Establish a hot asthma model: during the first two weeks of the experiment,Every of BCDE mice was fed 1g/ml of Codonopsis pilosula,huangqi decoction,0.57ml/d,two weeks later,the heat mice model was successfully estab-lished;In the first,seventh and fourteen days,mice were sensitized with ovalbumin solution,and to stimulate mice ovalbumin solution;themice were agitated and uneasy,Activity,head facial,Quiet,hair erect,severe cases,breast reduction,abdominal curl and incontinence.After the success of the induced asthma mice performance: active frequently,restlessness;Then bristling,less prone,respiration,depression;Moreover chest ups and downs,and neck,mainland incontinence,etc.Drug intervention experiment: Maxingxianxiongkeli group:since the first day of the inspired,feeding Maxingxianxiongkeli the original side daily,according to normal human dose 9.1 times,once a day,fed 0.36 for 1 week.;Dexamethasone group: since the first day of the in-spired,dexamethasone 3mg/kg/d was administered for 1 week.Maxing xianxiongkeli + dexamethasone group: since the first day of the inspired,the maxing xianxiong granules solution was given daily,once a day,0.36ml/20 g,and dexamethasone 3mg/kg/d for 1 week.In the 12 hours of the last stimulation,took the eyeball for blood,centrifuged serum,and d issected the Neuromicroscope lung tissue.The contents of-12,il and-13 i n serum of each group were determined and histopathological examination was done to give a view of the exudation and pathological changes in l ung tissue of mice.Results: Compared with the blank group,model control group IL–12levels were decreased significantly,with statistical significance(p<0.05);Compared with model group,the Maxingxianxiongkeli group,dexamethasone group,can increase the IL–12 at levels,with statistical significance(p<0.05);Compared to the model group,Maxing xianxiongkeli + dexamethasone group: IL–12 at the level of change is no differenceno statistical signific ance(p>0.05),increased significantly,the difference between the two group s(p<0.05),suggesting that maxing granules and Betamethasone may be tre ated asthma by raising serum il level.The dexamethasone group and Ma xingxianxiongkeli group had no significant difference in raising serum il l evel(p>0.05),suggesting that the two groups were effective.On the other hand,the change of serum IL-13 content is the opposite of the above chan ge trend..The lung tissue sections showed that: the lung tissues stained by hestaining were observed under light microscope: the bronchial mucosa of the blank group was smooth and intact,the pulmonary structure was clear,the inflammatory cells and inflammatory cells were not seen;In the model group,inflammatory cells were seen in the lumen and lung tissue,with inflammatory cell infiltration,necrosis of epithelial cells and large number of goblet cells,hyperemia of pulmonary blood vessels,bronchial cilia,with a certain number of adhesion,lodging and necrosis;The inflammatory infiltration and hyperemia in each group were milder than that in the model group.Conclusion: Using codonopsis,huangqi decoction and ovalbumin can successfully prepare mouse model of asthma.Maxing xianxiongkeli can raise IL-12 level,Down IL-13 level,it may be the treatment by adjusting the imbalance of TH1/TH2.
