| Objective: To observe the effect of maxingshuchuan Decoction on the expression of MUC5AC(mucin 5AC)and m CLCA3(calcium activated chloride channel type III)in asthmatic mice and to explore its mechanism,so as to provide experimental basis for further clinical application of maxingshuchuan decoction.Methods: 60 healthy female BALB / c mice were divided into 6 groups randomly:blank group,model group,low dose group,middle dose group,high dose group and dexamethasone group.The asthma model of mice was induced by ovalbumin(OVA).After the model was established successfully on the 14 th day of the experiment,30 minutes before each atomization of asthma,the treatment group and dexamethasone group were respectively given maxingshuchuan Decoction and dexamethasone by gavage,the other groups were given normal saline by gavage,and the samples were collected and detected after 7 days of administration.He staining was used to observe chronic airway inflammation and morphological changes,The proliferation of goblet cells in airway mucosa was observed by ab-pas staining,the contents of m CLCA3 and MUC5 AC protein in lung tissue of mice in each group were detected by Western blot,and the calcium activated chloride channel III was detected by RT-PCR The expression of type(m CLCA3)m RNA and MUC5 AC m RNA.Result:1.The morphological scores and airway inflammation of mice in the two groups were significantly better than those in the model group.2.The proliferation of goblet cells in dexamethasone group and maxingshuchuan Decoction group was less than that in model group.3.The expression of MUC5 AC and m CLCA3 protein in the model group was significantly higher than that in the blank group(P < 0.01);after dexamethasone and maxingshuchuan Decoction were used,compared with the model group,the content of m CLCA3 and MUC5 AC protein in the lung tissue decreased(P < 0.01 or P < 0.05).4.The expression level of MUC5 AC and m CLCA3 in the model group was significantly higher than that in the blank group(P < 0.01),and the expression level of m CLCA3 and muc5acmrna in the lung tissue of the model group was lower than that in the model group(P < 0.05 or P < 0.01).Conclusion:According to the results of this experiment,it is preliminarily determined that matingshuchuan decoction can improve the behavioral performance of mice,alleviate the symptoms of asthma,inhibit the inflammation of mice,reduce the proliferation of goblet cells and mucin MUC5 AC secretion.It is speculated that the mechanism is to reduce MUC5 AC protein secretion and control the gas to mucin high by inhibiting the proliferation of goblet cells,down regulating the expression of m CLCA3 gene and inhibiting the activity of m CLCA3 Secretion and inhibition of airway inflammation,relieve airway obstruction,reduce asthma symptoms... |
