A Cross-check Analysis Of Echocardiography,myocardial Injure Markers And Pathological Features In Rats With Alcoholic Cardiomyopathy

?Objective? By means of buliding a rodent model of ACM(alcoholic cardiomyopathy)to explore the relationship of echocardiography,myocardial injure markers and pathological features in the model rats.?Methods? 52 male SD rats were randomly divided into four groups,Alc+AT group(n=16),Alc+NS group(n=16),NS+AT group(n=10)and NS+NS group(n=10)respectively.All rats were given an adaptive intragastric administration for two weeks.Then,Rats from Alc+AT group were gavaged with alcohol,Simultaneously,the specific inhibitor of catalase activity,3-ammo-I,2,4-triazole,was injected intraperitoneally(1 g/kg body weight)every other day,and then let them drank 10%V/V alcohol at will.Alc+NS group were gavaged with alcohol,given normal saline intraperitoneally and 10%V/V alcohol drinking water.NS+AT group were gavaged with normal saline,given 3-ammo-I,2,4-triazole intraperitoneally and nomal drinking water.NS+NS group were gavaged with normal saline,given normal saline intraperitoneally and normal drinking water.After 10 weeks,the animal model was established.Echocardiography,myocardial injure markers and pathological features were examined in all rats.?Results? Myocardial injure markers(CK-MB,c Tn I)The study revealed that serum level of CK-MB,c Tn I in the Alc+AT group was obviously higher than which in other groups(p<0.05).Serum level of CK-MB,c Tn I in Alc+NS group was obviously higher than NS+AT group and NS+NS group,but lower than Alc+AT group(p<0.05).Echocardiography Compared with NS+AT group,IVSDd,LVPWDd,RWT,EF,FS,Em of rats from Alc+AT group were decreased(p<0.01),which even lower than its baseline value,while LVEDd,LVM,LVM/BW were increased(p<0.05).Compared with NS+NS group,Alc+NS group only had mild heart function change,such as EF,FS,Em,Em/Am decreased mildly(p<0.05).Compared with Alc+NS group,IVSDd,LVPWDd,RWT,EF,FS,Em,Am of rats from Alc+AT group were significantly decreased(p<0.01),while LVEDd,LVM/BW were increased(p<0.01).Compared with NS+NS group,NS+AT group only showed Em slightly lower(p<0.05).Compared with Alc+NS and NS+NS group,heart rate of Alc+AT and NS+AT group showed significant reduction(p<0.01),while there was no significant difference between Alc+NS and NS+NS group or between Alc+AT and NS+AT group(p>0.05).Histopathology Observations According to pathological results,the HE staining and transmission electron microscope in rats of NS+AT and NS+NS group were normal,while that of Alc+AT group rats were obvious abnormal,such as diffuse cardiomyocyte atrophy and diffuse myofilament dissolving.Compared with Alc+AT group,the histopathology changes of Alc+NS group were mildly and localized.?Conclusion? 3-ammo-I,2,4-triazole was beneficial to establish rats model of ACM.Myocardial injury markers,echocardiography results associated with pathological change shown in rats.Myocardial injury markers has advantage in early detection of myocardial injury.Echocardiography can accurately reflect the extent of myocardial damage in ACM rats.Myocardial injure markers and echocardiography were effective methods for researching ACM in rats.
