Analysis Of The Association Of Chronic Urticaria With Level Of CD4~+CD25~+ FOXP3~+T Cells,IL-10,IL-17,IL-23

Objective To investigate the expression of CD4 + CD25 + Foxp3 + Treg cells and IL-10,IL-17,IL-23 in the sera of patients with chronic urticaria.To discuss the CD4 + CD25 +Foxp3+Treg cells and Th17 cells,interleukin-17(IL-17),interleukin-23(IL-23),interleukin-10(IL-10)in the immunological mechanisms pathogenesis of chronic urticaria.Methods Make serums collection of 50 CU patients and 40 healthy control subjects,and they were all from the affiliated hospital of Qingdao university huangdao campus.The levels of CD4 + CD25 + Foxp3 + Treg cells were measured with Flow Cytometry.The expression of IL-10,IL-17,IL-23 were detected by ELISA.Set P < 0.05,the difference was statistically significant.The data comparison between the two groups using the t test and Pearson correlation analysis to make an statistical analysis.Results(1)In the peripheral blood CD4 + CD25 + Foxp3 + Treg cell levels(1.59 � 0.50)%of patients with chronic urticaria significantly lower than the healthy control group(3.24�0.77)%,the difference was statistically significant(P <0.05),and the course of the elder peripheral blood Treg cell levels(1.18� 0.33)% lower than the level of shorter duration(1.79�0.29)%,the difference was statistically significant(P <0.05).(2)In serum of patients with chronic urticaria the IL-10 levels(199.09 � 149.99)pg/ml slightly higher than the healthy control group(162.79� 134.44)pg/ml,there was no statistically significant difference(P > 0.05).(3)In serum of patients with chronic urticaria IL-17 levels(294.09�76.97)pg/ml significantly higher than the healthy control group(104.71�43.58)pg/ml,the difference was statistically significant(P < 0.05).(4)In serum of patients with chronic urticaria IL-23 levels(2189.13�612.49)pg/ml significantly higher than the healthy control group(962.20�383.92)pg/ml,the difference was statistically significant(P <0.05).(5)In serum of patients with chronic urticaria IL-17 levels and IL-23 levels were positively correlated(r = 0.439,P < 0.05).(6)In serum of patients with chronic urticaria IL-10 levels and CD4 + CD25 + Foxp3 + Treg levels were negatively correlated(r =-0.263,P > 0.05).(7)In serum of patients with chronic urticaria IL-17 levels and CD4+CD25+Foxp3 + Treg levels were negatively correlated(r = 0.264,P > 0.05).Conclusion The levels of CD4 + CD25 + Foxp3 + Treg cells and the expression of IL-17,IL-23 were abnormal in the patients with chronic urticaria.This result suggest that the occurrence of chronic urticaria is possible associated with autoimmune.Significance Through the study of the subject to provide experimental basis for the prevention and treatment of chronic urticaria,so as to provide new ideas and methods for the research and treatment of other allergic skin diseases.
