Coronary atherosclerotic heart disease is one of the major diseases that seriously threaten human life and health.In recent years,various advanced technologies(such as thrombolytic therapy,coronary rotational atherectomy,percutaneous coronary intervention and coronary artery bypass grafting)play an important role in opening blood vessel blockage,saving dying myocardium and reducing the myocardial infarction area and improving cardiac function,but the incidence of coronary heart disease is still increasing.Coronary heart disease not only increase the family and social economic burden,but also increases the risk of death.Therefore,prevention and treatment of coronary heart disease has become the focus of attention of the whole society.The pathogenesis of coronary heart disease is mainly due to coronary atherosclerotic plaque instability,in recent years,the study found that inflammatory reaction mediated by TLR4/NF-?B signaling pathway involved in the formation of unstable plaque,resulting in vascular occlusion,thrombosis,myocardial ischemic necrosis.At present,the main treatment of coronary heart disease is Western medicine.But some contraindications and side effects of combined medication lead to some patients with coronary heart disease can not long-term taking medicine.Guizhi Decoction prescription for treatment of coronary heart disease has thousands of years,effect curatively.The study is to investigate whether the protection of composite prescription Guizhi Decoction preconditioning against New Zealand rabbit myocardial ischemia is mediated by TLR4/NF-?B signaling pathway,and to provide theoretical basis for clinical prevention of myocardial ischemia.Objective:The study is to investigate whether the protection of composite prescription Guizhi Decoction preconditioning against New Zealand rabbit myocardial ischemia is mediated by TLR4/NF-?B signaling pathway,and to provide theoretical basis for clinical prevention of myocardial ischemia.Methods:1.Tirty-six New Zealand white rabbits were randomly divided into sham group,model group and drug group.Drug group with composite prescription Guizhi Decoction(6g/kg/d)gavage 20 days(prepared from affiliated teaching hospital preparation room of Hebei University of Engineering).Sham group and model group were given the same amount of distilled water for 20 days.The rabbit model was established by ligaturing the left anterior descending coronary artery(drug group and model group),sham group only thread ligation.2.Detecting cardiac ejection fraction of New Zealand rabbits after ligation by color Doppler echocardiography.3.The three groups of animals were extracted venous blood after ligation four hour,placed in the EP tube,static,centrifugation,stay in serum.Enzyme-linked immunosorbent assay(ELISA)was used to detect serum content of TNF-? and IL-6.4.After drawing blood,kill animals by injecting air into ear vein and take myocardial tissue by real-time quantitative reverse transcription polymerase chain reaction(RT-PCR)method for the detection of TLR4 m RNA expression,detect the expression of NF-?Bp65 by immunohistochemical method.5.The data was analyzed by SPSS 19.0 software.All data are presented by mean ± SD((X|-)±s).Multiple comparisons were performed with one-way ANOVA,two planned primary comparisons were performed with SNK-q.A value of P<0.05 was considered to denote statistical significance.Results:1.Before ligation,there was no significant changes in the electrocardio-gram of animals in each group.After ligation,the ST segment of ECG of model group and drug group was significantly higher.2.Before ligation,there was no significant changes the cardiac ejection fraction of animals in each group.After ligation,compared with sham group(71.32±2.49)%,the cardiac ejection fraction ofanimalsinmodelgroup(40.57±3.39)%and drug group(59.42±3.56)% were decreased(P<0.05);compared with the model group,the cardiac ejection fraction of animals in drug group was increased(P<0.05).3.The expression level of TNF-? and IL-6 in model group and drug group increased compared with sham group,but the drug group lower than model group(P<0.05).4.The expression of TLR4 m RNA in model group(3.45±0.28)and drug group(2.09±0.14)are higher than sham group(0.68±0.17),but the drug group lower than model group(P<0.05).5.The expression of NF-?Bp65 in model group(23.8±1.34)and drug group(14.7±1.15)are higher than sham group(3.5±0.28),but the drug group lower than model group(P<0.05).Conclusion:The above research implies that the composite prescription Guizhi Decoction can improve cardiac ejection fraction,cardiac function,increase myocardial tolerance,reduce myocardial injury and protect the damaged myocardium and its mechanism may be related to TLR4/NF-?B signaling pathway.
