Objective:To observe dynamic changes of serum Heart-type fatty acid-binding protein(H-FABP) in myocardial ischemia New Zealand rabbit and intervention effect of Guizhi Soup compound prescription, to study the protective effect of Guizhi Soup compound prescription on myocardium and to provide new ideas for clinical treatment and the development of safe and effective traditional Chinese medicine preparation.Methods:30 New Zealand rabbits with half males and half females,weighing 2.0 ~ 2.2kg, provided by the Experimental Animal Center of Hebei Medical University,were randomly divided into three group(nomal group,model group and drug group) using the random number table method, each group of 10.Drug group with guizhi Soup compound prescription(6g/kg/d) gavage 20 days(prepared from affiliated teaching hospital preparation room of Hebei University of Engineering). normal group and model group were given the same amount of normal saline for 20 days.The myocardial ischemia model was created by left anterior descending coronary artery ligation(drug and model groups), wearing the normal group without ligation. Three groups respectively before ligation, ligation after 0.5h, 1h, 2h, 4h, 6h obtained venous blood,centrifuged and serum specimens spare. Enzyme-linked immunosorbent assay(ELISA) was used to detect serum content of H-FABP. Color doppler echocardiography(CDE)computed ejection fraction(EF).The data was analyzed by SPSS 19.0 software.measurement data use ?x± s representation. Comparison between the groups and within the groups used the analysis of variance and SNK-q test. a time-dependent variable used the repeated measures analysis of variance.Difference was considered to be statistically significant at P<0.05.Results:1. normal group, model group and drug group the content of serum H-FABP with myocardial ischemia time all have dynamic change.And after ligation 0.5h serum H-FABP content began to increase, 4h reached a peak, 6h began to decline, P <0.05, the difference was statistically significant. The model group 0.5h, 4h and 6h the content of serum H-FABP were 2.17 ± 0.25ng/ml, 3.66 ± 0.30ng/ ml and 2.22 ± 0.21ng/ml. 2. Before ligation, The normal group, model group and drug group H- FABP content in serum were 1.05±0.19ng/ml,1.16±0.38ng/ml and 1.00±0.17ng/ml, there was no statistically significant difference(P > 0.05).3. After ligation, compared with normal group, model group and drug group of each time point the H- FABP content in serum were increase,P < 0.05,the difference was statistically significant;Compared with model group, drug group of each point in time the content of serum H- FABP is reduced,P < 0.05,there was statistically significant difference.4. Before ligation, the heart ejection fraction of the normal group, model group and drug group of were 72.32±2.99%,73.12±2.12% and 71.56±2.57%,the difference was not statistically significant(P> 0.05).5. After ligation 6h, compared with the normal group,model group and drug group,the cardiac ejection fraction was decreased,P <0.05,the difference was statistically significant; compared with the model group, the cardiac ejection fraction drug group was increased,P <0.05,there was statistically significant difference.Conclusion:1. the content of serum H- FABP have dynamic changes in myocardial ischemia New Zealand rabbit.And after myocardial ischemia 0.5h the content of serum H-FABP began to increase, 4h reached a peak, 6h began to decline.H-FABP can early evaluation of myocardial injury, is an early biochemical marker of myocardial injury. Therefore, in the early myocardial ischemia monitorring the content of serum H- FABP can quickly identify high-risk patients, earlyier take reasonable treatment measures, improve the prognosis and the survival rate of patients.2. The preventive application of guizhi Soup compound prescription in New Zealand rabbit,when not subject to cardiac ischemic injury, had no effect on the content of serum H-FABP and the cardiac ejection fraction; and when suffered the myocardial ischemia, can reduce the levels of serum H-FABP and improve cardiac ejection fraction, improving heart function.
