Objective:By the classic pharmacodynamic experiment of transdermal drug preparation,Studying the analgesic and anti-inflammatory effects of XiaoZhongDingTongtie in the treatment of acute soft tissue injury.By observing the effect of the drug on serum PGE2 and IL-1β contents in animals,and to explore the mechanism of action.By observing the sensitivities of the drug on the back model of guinea pigs,the safety of skin medication was verified,which provided scientific basis for clinical use.Method:Reference modeling method reported,the model of acute closed soft tissue injury was established according to the best impact condition explored by the pre-experiment.Choosing Qi ZhengXiaoTongtie as the control group,to observe the effect of XiaoZhongDingTongtie on the repair of damaged tissue by comparing the injury score at different time points.Respectively,with writhing and hot plate experiment to study its analgesic effect,with ear swelling and foot swelling test to study its anti-inflammatory effect.The levels of PGE2 and IL-1β in the serum of rats were determined by enzyme-linked immunosorbent assay(ELISA)which was used to illustrate the therapeutic action of XiaoZhongDingTongtie,it could inhibit the release of inflammatory mediators.Through the guinea pig back skin drug sensitization experiments to observe the safety of XiaoZhongDingTongtie.Result:(1)Injury syndrome index: The high,medium and low dose groups could improve the local swelling and ecchymosis,and the difference was statistically significant compare with model group(p<0.05).(2)Analgesic effect:The high and middle dose groups could significantly inhibit the number of wrinkles in mice induced by acetic acid(p<0.05);Each dose group can improve the pain threshold of thermal stimulation in mice(p<0.05),high and medium dose group compared with the QiZheng group,the thermal stimulation pain threshold was no significant difference(p>0.05).(3)Anti-inflammatory effects:High,medium and low dose groups can inhibit xylene-induced ear swelling in mice,compared with the blank group,the difference was statistically significant(p<0.05),the inhibitory effect and QiZheng group was no significant difference(p>0.05);The high,medium and low dose groups could inhibit the swelling of the egg in rats(p<0.05),and there was no significant difference between thehigh and middle dose groups and the QiZheng group(p>0.05).(4)Inflammatory mediators:The release of PGE2 was inhibited at 3d and 7d in the high and middle dose groups(p<0.05),and the release of PGE2 was inhibited on the 7th day in the low dose group(p<0.05);In the high dose group,the inhibition of PGE2 on the 3rd day was no significant difference compared with the QiZheng group(p>0.05),the inhibition of PGE2 on the 7th day in the high,middle and low dose groups was not statistically significant compared with the odd group(p>0.05).The inhibition of IL-1β in the high,medium and low dose groups was statistically significant compared with the model group on the 3rd and 7th day(p<0.05),there was no significant difference between the high and middle dose groups on the 3rd and 7th day compared with the Qi Zheng group(p>0.05),there was significant difference between the low dose group and the QiZheng group on the 3rd day and the 7th day(p<0.05).XiaoZhongDingTongtie did not cause guinea pig skin allergies.Conclusion:XiaoZhongDingTongtie has obvious analgesic and antiinflammatory effects,and has obvious repair effect on acute closed soft tissue injury,reducing the content of PGE2 and IL-1β in serum may be one of the mechanisms and short-term use no obvious skin allergies.
