Objective: To observe the effect of Chinese medicine compound malt pill(CMP)on estrus cycles and ovary morphology changes,expression of Follicle-stimulating hormone(FSH),Luteinizing hormone(LH),3?-hydroxysteroid dehydrogenase(3?-HSD)and cytochrome P-450 enzyme system(CYP450)in rat model of polycystic ovary syndrome(PCOS),and to further explore its possible mechanism and lay a clinical foundation.Method: 60 female Sprague-Dawley rats aged 9 days were randomly divided into normal control group(n=10)and model group(n=50).Model group rats were injected subcutaneously with testosterone propionate for inducing PCOS model.The successful model rats were randomly divided into 5 groups: model group,positive control group,and high-,moderateand low-dosage CMP group(n=9).The treatment groups were given intragastric administration with corresponding drugs,the normal group and the model group were given distilled water.All rats were lavaged for 21 days continuously.The changes of estrous cycle were observed by Wright staining,ovarian morphology change were observed by HE staining,and expression of FSH,LH,3?-HSD and CYP450 were detected by immunohistochemical,western blot,reverse transcription PCR or quantitative real-time PCR after treatment.Results:(1)The estrus cycles of PCOS rats showed irregular change and get right partly with drug intervention.(2)The cystic dilatation of follicles increased and granulosa cell decreased in model group.After treatment,the diameter of cystic follicles decreased in different degrees,granulosa cell and corpus luteum increased.(3)The expression of LH were significantly increased and FSH were significantly decreased in model group than that in normal group(P<0.01),compared with model group,the expression of LH were significantly decreased and FSH were significantly increased in CMP each dose group(P<0.01).(4)The expression of 3?-HSD were significantly increased and CYP450 were significantly decreased in model group than that in normal group(P<0.01),compared with model group,the expression of 3?-HSD were significantly decreased and CYP450 were significantly increased in CMP each dose group(P<0.01).Conclusion: The treatment of PCOS with compound malt pill may be realized by improving the estrous cycle disorder,polycystic ovarian morphology and regulating the expression of FSH,LH,3?-HSD,CYP450 in ovarian tissue.
