OBJECTIVE: Our aim was to assess the correlation of serum sclerostin with coronary atherosclerotic heart disease(CHD)and osteoporosis.METHODS: Three hundreds and thirteen elderly patients who underwent coronary angiography(CA)were involved in the cross-sectional study,which included 163 subjects in CHD group and 150 subjects in non-CHD group.When further stratifying the subjects according to the result of DXA,there are 163 osteoporosis patients and 150non-osteoporosis patients.Serum sclerostin concentrations were measured using a commercially available enzyme-linked immunosorbent assay kit.The levels of factors of cardiovascular disease and bone turnover markers were measured in serum.The stenosis of coronary artery was detected by coronary angiography.A stepwise multi-linear regression analysis was used to determine the factors which independently correlated with sclerostin.The association of sclerostin and CHD was investigated by multiple logistic regression.RESULTS: Serum sclerostin concentrations were lower in elderly patients in CHD group than patients in non-CHD group[(178.035 ±0.636)pg/m Lvs(180.576±0.608)pg/m L,P<0.05],and the significance was maintained in osteoporotic patients[(177.4+7.1)pg/m Lvs(180.1+3.4)pg/m L,P<0.05] rather than non-osteoporotic patients.Age(?=-0.162,P<0.05)was negatively correlated with sclerostin,bone mineral density(BMD)(?=0.224,P<0.05)and PINP(?=0.161,P<0.05)were positively correlated with sclerostin.In the multiple regression analysis,sclerostin concentrations were one of the independent factors that remained significantly associated with CHD both for the whole patients(OR=0.865,P=0.018)and the osteoporotic patients(OR=0.767,P=0.007).CONCLUSIONS: Circulating sclerostin was decreased in elderly CHD patients,and could be a major modulator of Wnt signaling in CHD with implications in osteoporotic patients.
