Objective:To explore the effect of Addie injection on the immune system of rabbit tongue squamous cell carcinoma(cell line VX-2)model.Methods: 48 rabbit tongue squamous cell carcinoma models and healthy rabbits(n = 24)were selected and randomly divided into 6 groups.Group A(healthy rabbit + saline),Group B(healthy rabbit + Addie injection),Group C(tumor-bearing rabbit + saline),Group D(tumor-bearing rabbit + Paclitaxel),Group E(tumor-bearing rabbit + Addie injection),Group F(tumor-bearing rabbit + Addie injection + Paclitaxel).Drug was injectedvia the rabbit marginal ear vein.Peripheral blood was collected 1 daybefore drug administration as well as7 and 14 daysafterward.Changes of IL-2,IL-6,IL-10,IFNand TNFin peripheral blood were measured by Elisa.6 rabbits in each group were euthanised on the 7th day and the 14 th day.Spleen was harvested and spleen indexcalculated.HE staining was used to observe the morphological characteristics of primary lesions.SPSS wasused for statistical analysis.Results: 1.Tumor formation rate onmodel animal was 93.33%.HE staining of the lesions showed that the structure of tumor tissue is in disorder,with low differentiated celland abnormal mitosis.2.Comparison between healthy rabbits:There was no significant difference in Elisa result between Group A and Group B before and after drug administration(P > 0.05).3.Comparison between tumor-bearing rabbits:There was no significant statistical difference in each group before and after the treatment(P > 0.05).Compared to Group C,D,E,the concentration of IFN-?,TNF-? and IL-2 of Group F were increased at the7 th and 14 th daysafter administration,and the concentrations of IL-10 and IL-6 were decreased.The difference between Group F and Group C and D was statistically significant(P < 0.05).There was no significant difference between Group F and Group E in IL-10 and IL-6 at the 7th and the 14 th days(P > 0.05).There was no significant difference in IL-2,IFN-?, and TNF-?(P < 0.05)at the 7th days,but there was significant difference between the two at the 14th(P < 0.05).Compared to Group C and D,the concentrations of IFN-?,TNF-? and IL-2 in Group E were increased at the 7th and the 14 th days of administration,and the concentrations of IL-10 and IL-6 were decreased.Among them,Group E and Group C showed significant difference in IFN-?,TNF-?,IL-10 and IL-6(P < 0.05),while the difference of IL-2 was not significantly different at the 7th days(P > 0.05),there was significant difference between the two at the 14th(P < 0.05).There was no significant difference between Group E and Group D in IFN-?,TNF-?,and IL-10 at the 7th(P <.0.05),but showed significant differences at the 14th(P < 0.05).There were significant differences in IL-2 and IL-6 at the 7th days(P < 0.05),but showed no significant difference at the 14th(P < 0.05).Comparedto Group C,the concentrations of IFN-?,TNF-? and IL-2 of Group D show littledifference at the 7th and the 14 th days,the concentrations of IL-10 and IL-6 were decreased,but was not statistically meaningful(P > 0.05).4.Comparison between healthy rabbits and tumor-bearing rabbits:There was no significant statistical difference between the Groups before the administration(P > 0.05),The concentrations of group A and group C showed A decrease in the concentration of IFN-?,TNF-?,and IL-2 in the seventh and 14 th days of the drug,and the concentration of IL-10 and IL-6 on the 14 th day of the drug increased without significant difference(P > 0.05),Comparedto Group B,the IFN-?,IL-2,TNF-? level of Group E were increased,IL-10,IL-6 were decreased at the 7th and 14 th days,however,only the difference of IL-10 at the 7th and 14 th,IL-6,IFN-? at the 14 thwere statistically meaningful(P < 0.05)while other difference were not(P > 0.05).5.The spleen index of Group E and Group F increased at the 7th and 14 th days of administration,and were statistically meaningful comparedto Group C and D(P < 0.05).The spleen index of Group E was higher than that of Group B(P < 0.05),Group B>Group A,Group A> Group C,Group D>Group C inspleen index,but was notstatistically significant(P > 0.05).Conclusion: Addie injection can effectively regulate Th1 and Th2 type cytokines and increase the spleen index in tumor-bearing rabbits,and is able to achieve the same effect when combined with Paclitaxel in chemotherapy.The balance of Th1/Th2 was shifted in favor of Th1,and the dominancy of Th1 was even moreprominentin combination chemotherapy.
