Effects Of Aidi Injection And Paclitaxel Injection Combined Chemotherapy On Angigenesis And Lymph Metastasis In Rabbit Tongue VX-2 Squamous Cell Carcinoma Model

Objective: To investigate the effects on tumor angiogenesis,VEGF expressing and lymphatic metastasis in rabbit tongue squamous cell carcinoma by single using Aidi injection and Paclitaxel injection combined chemotherapy.Methods:(1)48 rabbit tongue VX-2 squamous cell carcinoma models were established by surgical implantation and divided into 4 groups.24 healthy new zealand rabbits were randomly divided into 2 groups.The grouping design is listed as follows: group A(healthy rabbits + normal saline),group B(healthy rabbits + Aidi injection),group C(cancer xenograft model + normal saline),group D(cancer xenograft model + Aidi injection),group E(cancer xenograft model + Paclitaxel injection),group F(cancer xenograft model+ Aidi injection combined with Paclitaxel injection).The peripheral blood samples of rabbits were collected before the experiment,and all experimental animals were given medication via ear vein.(2)After the 1st and 2nd week of treatment,all peripheral blood samples were collected,then 6 randomly choosed rabbits of each group were killed and all primary tongue cancer samples and cervical lymph nodes were collected in preparation for further tests.(3)The pathological features of cancer and lymph nodes samples were observed by hematoxylin-eosin staining,the number of all lymph nodes and metastatic lymphadens were counted to evaluate the lymph node ratio of each xenograft model.(4)The serum VEGF concentration of all rabbits were detected by ELISA.(5)The density of micro blood-vessels,lymph-vessels and the VEGF expression level in tongue cancers were measured by immunohistochemical staining.(6)All data was analyzed by SPSS 23.0.Results:(1)The serum VEGF concentration: no statistical difference was found in serum VEGF concentration between group A and group B by each period of medication(P >0.05).There was no statistical difference in serum VEGF concentration among all experimental groups before the medication.After the 1st week of treatment,the serum VEGF concentration of cancer xenograft model group was higher than that of healthy rabbit group(P<0.05),while there was no statistical difference in serum VEGF concentration among cancer groups(P>0.05).And after the 2nd week of medication,the serum VEGF concentration of cancer xenograft model group was higher than that of healthy rabbit group(P<0.05),and the serum VEGF level was significantly lower in D,E,F group than C group(P<0.05),while there existed no statistical difference among D,E,F group(P>0.05).(2)The VEGF level in primary tongue cancer: the VEGF level of D,E,F group was lower after the 2nd week therapy than that after the 1st week(P<0.05).After the 1st week,the VEGF level of C,D,E,F group had no statistical difference(P>0.05).After the2 nd week,the VEGF level of D,E,F group was lower than that of group C,and the level of E,F group was lower than group D(P<0.05),but no statistical difference was found between group E and F(P>0.05).By using the Pearson correlation analysis,the serum VEGF concentration was positively correlated with that in primary tongue cancer(r=0.592,P<0.01).(3)The micro vessel density of tongue and primary cancer: there was no statistical difference in MVD of tongue between group A and group B by each period of medication(P>0.05),and the MVD of D,E,F group was lower after the 2nd week therapy than that after the 1st week(P<0.05).After the 1st week of medication,no statistical difference was found in MVD among C,D,E,F group.While after the 2nd week,the MVD of D,E,F group was significantly lower than that of C group(P<0.05),and the MVD of E,F group was lower than D group,but there was no statistical difference being found between these two groups.By using the Pearson correlation analysis,the MVD was positively correlated with the VEGF level expressed in tumor tissue(r=0.770,P<0.01).(4)The micro lymph-vessel density of tongue and primary cancer: no statistical difference was found in MLVD between group A and group B by each period of medication(P>0.05).The MLVD of E,F group was lower after the 2nd week therapy than that after the 1st week(P<0.05).No statistical difference in MLVD was found among C,D,E,F group after the 1st treating week.After the 2nd week of treatment,the MVD of group E,F was lower than that of group C,D(P<0.05),but no statistical difference was found between the two groups(P>0.05).(5)There was no significant difference found in lymph node ratio among C,D,E,F group after the 1st and 2nd week of medication(P>0.05).Conclusion: Aidi injection can reduce angiogenesis and VEGF expressing in rabbit tongue VX-2 squamous cell carcinoma model,but have no significant influence on lymphangiogenesis and cervical lymph node metastasis.Also Aidi injection cannot improve the efficacy of Paclitaxel injection chemotherapy in reducing angiogenesis,lymphangiogenesis and VEGF expressing.
