Objective:To investigate the influence of different ways chemotherapy on apoptosis ofmetastatic lymph node cancer cells in rabbits VX-2squamous cell carcinoma of tongue.Methods:A total of24New Zealand rabbits with tongue implantation of VX-2tumorswere divided into6groups randomly,with4in each.Chemotherapy was carried out oncelymphonodi cervicales reached5mm in the maximum transverse diameter.Group Areceived a lingual artery perfusion of paclitaxel liposome and carboplatin injection.GroupB received a lingual artery perfusion of free paclitaxel injection and carboplatininjection.Group C received a lingual artery perfusion of5%glucose injection.Group Dreceived a auricular venous perfusion of paclitaxel liposome and carboplatininjection.Group E received a auricular venous perfusion of free paclitaxel injection andcarboplatin injection.Group F received a auricular venous perfusion of5%glucoseinjection.The dose of paclitaxel and carboplatin in each administration respectively was7.5mg/kg and13.0mg/kg.lymphonodi cervicales were resected and tested after One week.①Transmission electron microscope was used to observe the changes of the microstructureof apoptotic metastatic cells.②Flow cytometry were used to tested the changes of theapoptosis rate of metastatic cells, The significance of the differences was determined withthe one-way ANOVA followed by the Fischer LSD test.③Expressions of P53protein wereexamined by using the SP immunohistochemical method,Chi-square test and Fisher’s exacttest were used for numeration data.Results:①T he treatment group showed obvious apoptosis of cancer cells.②The group ofcancer cells apoptosis rate of intra-arterial chemotherapy was higher than that ofintra-venous chemotherapy group. The group of cancer cells apoptosis rate of paclitaxelliposome and carboplatin injection was higher than that of free paclitaxel injection andcarboplatin injection group.Goup A lymph node shrink,the rests without shrinking.Theapoptosis rate in groups A,B,C,D,E and F were (49.49±4.96),(29.65±3.61),(8.59±0.28),(21.05±1.65),(12.98±0.75) and (8.47±0.30)%,respectively.There were significant differences between compared to the rest of the group sets (P<0.05for both) except between group Cand group F.③The positive rates of P53expressions in group A,B,D,E,C and F were20.00%(2/12),33.33%(4/12),41.67%(5/12),50%(6/12),and double75%(9/12),respectively.There were significant differences between group A and group C and group F (P<0.05),there were no statistical difference between two other groups.Conclusion:Intra-arterial chemotherapy was better in induces apoptosis of metastatictumor cells and inhibits the proliferation of tumor cells in lymph node thanintra-venous.Paclitaxel liposome and carboplatin injection by intra-arterial chemotherapycan effectively induces apoptosis of metastatic tumor cells and inhibits the proliferation oftumor cells in regional lymph node.
