Effects Of Aidi Injection Combined With Paclitaxel Chemotherapy On Regulatory T Cells Of VX-2 Rabbit Tongue Squamous Cell Carcinoma Model

Object: To investigate the effects of Aidi injection combined with paclitaxel injection chemotherapy on regulatory T cells,cytokines IL-10,IL-35 and TGF-? of VX-2 squamous cell carcinoma model of rabbit tongue.Methods: Forty-eight rabbits with tongue cancer were randomly divided into four groups,and twenty-four healthy rabbits were randomly divided into two groups.A total of six groups: group A(healthy rabbits + normal saline),group B(healthy rabbits + Aidi injection),group C(tumor rabbits + normal saline),group D(tumor rabbits + paclitaxel injection),group E(tumor rabbits + Aidi injection),group F(tumor rabbits + paclitaxel injection + Aidi injection).Experimental rabbits were injected with medicine via ear vein after 15 day following the tumor rabbits were established.Firstly,on the 14 th,22th and 29 th day after all tumor models were established,we collected peripheral blood samples.On the 22 th day,six rabbits were randomly killed from each group and the samples were collected,then the rest of rabbits were given the same treatment on the 29 th day.The histomorphological features of rabbit tongues were detected by HE staining.The percentage of CD4+CD25+CD127low Treg/CD4+T cells in peripheral blood,cervical lymph nodes,primary tumor of tongue cancer and peritumoral cancer were detected by FCM.The serum levels of IL-10,IL-35 and TGF-? were detected by ELISA.Results:(1)The percentage of Treg cells in peripheral blood and cervical lymph nodes as well as the concentrations of IL-10,IL-35 and TGF-? in serum of group B were not significantly different from that of group A by each time period of treatment(P>0.05).(2)Before the medication,the percentage of Treg cells in peripheral blood and the serum concentration of IL-35 were higher in cancer groups than group A(P<0.05).After the 1st week of treatment,the percentage of Treg cells in peripheral blood,the serum concentration of IL-35 and TGF-? were both higher in cancer groups than groupA(P<0.05),while the percentage of Treg cells in lymph nodes of C,D group was higher than that of group A(P<0.05),and the serum concentration of IL-10 of A group was lower than that of group C,D,E(P<0.05).After the 2nd week of medication,the percentage of Treg cells in peripheral blood as well as the serum concentration of IL-35 were higher in cancer groups than A group(P<0.05),the percentage of Treg cells in lymph nodes and the serum expression level of TGF-? were higher in C,D,E group than A group(P<0.05),and the IL-10 level was higher in C,D group than group A(P<0.05).(3)The percentage of Treg cells in peripheral blood and cervical lymph nodes as well as the concentrations of IL-10,IL-35 and TGF-? in serum of group D,E,F were lower than that of group C after the 1st week of treatment(P<0.05).And no significant differences were found in indexes above among the D,E and F group(P>0.05).The concentration of IL-10 in serum of group D was lower than that of group C(P<0.05).There was no significant difference in the percentage of Treg cells in primary tongue cancer between group E and C(P>0.05),while other indexes of group E were lower than that of group C(P<0.05).The percentage of Treg cells in cervical lymph nodes and the concentration of IL-10 in serum of group E were lower than that of group D(P<0.05).Indexes mentioned above of group F were lower than that of group C(P<0.05).No significant differences was found in the percentage of Treg cells in primary tongue cancer between group F and group D(P>0.05),while other indexes of group F were lower than that of group D(P<0.05).The concentration of TGF-? in serum of group F was lower than that of group E(P<0.05).(4)The percentage of Treg cells in cervical lymph nodes and peritumoral tissue after the 2nd week of treatment was higher than after the 1st week in group C,D(P<0.05).There was no significant difference in the percentage of Treg cells in primary tongue cancer by each time period of treatment among group C,D,E and F(P>0.05).(5)The concentrations of IL-10,IL-35 and TGF-? in serum were positively correlated with the percentage of Treg cells in peripheral blood of experimental rabbits(0.5<r<1,P<0.01).Conclusion: Aidi injection combined with paclitaxel injection chemotherapy can reduce the percentage of CD4+CD25+CD127low Treg/CD4+T cells in peripheral blood,cervicallymph nodes,primary tongue cancer,peritumoral tissue and the concentrations of IL-10,IL-35 and TGF-? in serum of VX-2 rabbit tongue squamous cell carcinoma model.
