Objectives The study on treatment of chronic unpredictable mild stress(CUMS)and living alone in rats to build depression model rats,observing the effects of result on Shuxintang(SXT)on expression of behavior,hypothalamus-pituitary-adrenal axis(HPAA)and inflammatory factor interleukin 1?(IL-1?),interleukin 6(IL-6).Exploring the treatment of SXT on depression model rats.According to regulating the qi of five zangorgans theory of TCM,this paper discusses the antidepressant mechanism of SXT.So as to provide new method for clinical application of treating depression.Methods In living alone and CUMS to build depression rats' model.Before and after the experiment respectively measure the results of the rats' weight growth and sugar water consumption,open-field test,forced swimming test,and weight is determined once a week,which assesses the treatment of SXT to living alone and CUMS depression model rats' behavior.According to the ELISA kit specification to measure rats' serum adreno cortico tropic hormone(ACTH)and cortisol(CORT),interleukin 6(IL-6)and interleukin 1?(IL-1?)levels.Detecting the expression of CRHm RNA in hypothalamus and GRm RNA in hippocampus by RT-PCR.All the experimental data were expressed as a mean standard deviation((?) ±s),and the comparison between groups using single factor analysis of variance,with statistical significance(P ? 0.05),with obvious statistical significance(P?0.01).Results 1 The rats living alone and accepting CUMS after 28 days,model group rats' behavioral indexes of the rats were significantly decreased(P?0.05 or P?0.01)compared with the blank group with statistical significance.The model group rats' serum ACTH,CORT,IL-6,IL-1?,CRHm RNA in hypothalamus were significantly increased(P?0.05 or P?0.01)compared with the blank group with statistical significance,GRm RNA in hippocampus were significantly decreased(P?0.05 or P?0.01)compared with the blank group with statistical significance.2 The treatment by medicines after 28 days,SXT group,sertraline hydrochloride group and the two medicines combination group rats' behavioral indexes of the rats were significantly increased(P?0.05 or P?0.01)compared with the model group with statistical significance.The SXT group,sertraline hydrochloride group and the two medicines combination group rats' serumACTH,CORT,IL-6,IL-1?,CRHm RNA in hypothalamus were significantly decreased(P?0.05 or P?0.01)compared with the model group with statistical significance.GRm RNA in hippocampus were significantly increased(P?0.05 or P?0.01)compared with the model group with statistical significance.Conclusions 1 SXT can effectively improve the state of depressive rats and its behavior of the growth of body weight,the sugar water consumption rate,the vertical and horizontal movement score of open-field test,the time of forced swimming test.2 SXT can reduce levels of depressive rats' serum CORT and ACTH,IL-1?and IL-6,CRHm RNA in hypothalamus,and improve the level of GRm RNA in hippocampus.3 The mechanism of SXT may be related to adjust HPA axis and inflammation factor IL-6 and IL-1?.
