Objective:Study the liver flavored four inverse scattering the formulas of chronic stress depression model rats of HPA axis,nervous biochemical and the influence of related parameters of the autonomic nervous system,explore the liver factor s clear and smooth implementation of modern function and mechanism.Methods:1.Chronic mild stress of depression model rats,prepared to field experiment,the sugar water preference,cross a maze experiment,forced swimming test index evaluation model.Will stress group rats were divided into control group,the group(CMS),the positive drug group-fluoxetine group(group F),flavored group si-ni-san group(JS),spleen decoction group(group G),four inverse scattering group(group S),a total of six groups.2.Testing stress autonomic nervous function in rats.Using abdominal cavity anesthesia method,the use of common carotid artery intubation,electrocardiogram(ecg),methods of PowerLab system,records in the rat heart rate,heart rate variation parameter HRV,autonomic nerve indexes such as blood pressure3.Testing stress function of HPA axis in rats.ELISA test rats(after autonomic nerve function tests executed)trunk CRH,;CORT and ACTH levels in serum.4.Testing stress single amine neurotransmitter function in rats.ELISA to detect the rat hypothalamus,hippocampus and frontal cortex,5-HT,NE,DA contentResults:1,the rat model of evaluation:(1)Open field experiment OFT: experimental rats for 14 days,compared with CMS group,the rest of the five groups does not affect the total movement distance in OFT.After 35 days,compared with CMS group,group F OFT movement in the total distance increased significantly(p < 0.05),total JS group and the group S OFT movement distance increased significantly(p < 0.05),G group there was no significant difference.Experimental rats for two weeks,compared with CMS group,the rest of the five groups had no obvious effect on number OFT upright.After the stress,compared with CMS group,group F number OFT upright,JS group was obviously increased(p < 0.05),two groups of efficacy difference is not obvious.JS group make rats upright number increased significantly(p < 0.01),G group no significant difference.(2)preference experiments: sugar water stress after 14 days,between the CMS and other group rats sugar water preference degree of no significant difference.Sugar water preference coefficient after stress,stress group than the control group significantly reduced(p < 0.05),F,JS group significant rise in the stress group(p < 0.05),and other groups have no significant difference.(3)cross maze experiment,the effect of OE % in the EPM,after the stress,compared with the control group,the CMS group had significant difference(p < 0.05).JS group OE % more stress have improved,but without statistical significance.The effect of OT % in the EPM,after the stress,compared with the control group,the CMS group had significant difference(p < 0.05).OT % JS group significantly increased(P < 0.01).The rest of the group had no significant difference.Forced swimming(FST)experiment of(5): after the stress,FST suspension fixed number of CMS group have significant differences(p < 0.05),F,JS group after the intervention dropped significantly in the stress group(p < 0.05).6 rats body quality of growth: building the second week,groups of experimental rats weight increase,the CMS group compared with other groups growth is slow,but there was no significant difference between groups.After the stress,weight growth rate compared with control group,the CMS group was obviously lower(p < 0.0001)under the JS group,S body quality growth were significantly increased after the intervention group was given(p < 0.01).Conclusion:Chronic stress-induced depression model in rats with stable and reliable,modified liver recipe JWSNS can significantly improve the behavior of chronic stress depression model rats,the relative indexes of HPA axis,neurobiology and autonomic nervous system showed the overall adjustment of depression,and partly revealed the mechanism of the function of the main release of the liver to control the depressive mood state.
