Hyperuricemia is a disease caused by gene and abnormal pathway of regulating uric acid,resulting in the accumulation and secretion of uric acid produced by purine metabolic disorders.Hyperuricemia is not only the basis of the incidence of gout,but is also related to diabetes,high blood pressure and other cardiovascular diseases which is a serious threat to human health.In recent years,the number of patients with hyperuricemia is increasing,although western medicine has faster efficacy to reduce uric acid,it may cause a series of side effects.Natural products which can reduce uric acid are rich in resources and have a huge development potential and market prospects.However,there are still variety of problems in the current research,such as unstable screening model in vivo,unclear reducing-uric-acid active ingredients and etc.In this study,a stable screening model of acute hyperuricemia was established.The traditional Chinese medicine composition for the treatment of hyperuricemia was screened out by homogeneous design method and pharmacodynamic analysis.The pharmacological mechanism and preparation technology were studied and discussed.This research effectively solve the problems existing in natural reducing uric acid drug research and development and provide an effective basis for the development of anti-gout drugs.The full text is divided into the following six partsPart? Screening of Traditional Chinese Medicine Prescriptions Based on Uniform DesignOn the basis of previous experimental research,this chapter will apply the mathematical model to the research of Chinese medicine,follow the concept of compatibility of traditional Chinese medicine compound and play the advantages of traditional Chinese medicine compound multi-way pharmacological effects.Using the uniform design method,the efficacy of the evaluation index for the experimental study to explore the best combination ratio of drugs.The results showed that the optimal combination of the traditional Chinese medicine composition was the alcohol extract of Alpinia officinalis,Alpinia officinalis,Dandelion,Cichanthus japonicus and Psyllium,the best ratio was 10: 8: 7: 6:10.In addition,the mechanism of optimal prescription was investigated.It was found that the optimal prescription compatibility could effectively reduce the serum uric acid by inhibiting the activity of XOD in hyperuricemia mice and promoting uric acid excretion.Part? Study on the preparation technology of Chinese herbal compoundIn this chapter,the effective parts of the five medicinal herbs were used as raw materials.Mild,persistent pellet formulations were selected according to the basic principle of the selection of Chinese medicine dosage forms.The preparation method was used to optimize the process parameters of the extruded spheronization pellets.The optimum extrusion speed,sphericity and spheronization times were determined to be 40 RPM,35 RPM and 3 min,respectively.Finally,uric acid pellets were evaluated in vitro from pellet roundness,yield and stability.The results showed that the pellets had a roundness of(16.78 ± 0.57)° and a yield of(71.55 ± 1.08)%.The yield was good and the difference between the batches was small,which was in accordance with the relevant preparation requirements.Part ? Determination of the Active Ingredient of Reducing Acidic Pellets and Its PharmacodynamicsIn this chapter,the flavonoids and crude polysaccharides in the pellets were qualitatively analyzed.Then,the content of total flavonoids in uric acid pellets was determined by UV spectrophotometry using rutin as comparison sample.The analytical method of phenol-sulfuric was established to determine the crude polysaccharide content in uric acid pellets.Finally,the uric acid effect of the pellet formulation was evaluated.The results showed that the total flavonoid content in the pellets was about 10%,the crude polysaccharide content was about 1.94%.The quality standards of the reducing uric acid pellets to treat gout were established.Uric acid pellets had a significant reduction in uric acid(P < 0.01)...
