Objective:Through making acute and chronic hyperuricemia model, studying the effect and the mechanism of the total coumarin of Cortex Fraxini(TCCF) on the level of uric acid in hyperuricemia mice.Methods:1. Different dosage of Potassium Oxonate(ip.) was used for making acute hyperuricemia model of mice.To investigate the best dosage of potassium oxonate for modeling. Xanthine+Potassium Oxonate, Adenine+Potassium Oxonate, Adenine+Ethambutol and Xanthine+Ethambutol were used to investigate the best way for making chronic hyperuricemia model of mice..2. TCCF were administrated to acute hyperuricemia mice induced by potassium oxonate. Then observing the level of BUA, XOD, ADH, GDH in hyperuricemia mice.3. TCCF were administrated to chronic hyperuricemia mice. Then observing the level of BUA, XOD, UUA, CR, BUN in hyperuricemia mice.Results:1. Potassium Oxonate can obviously increase the level of BUA, but it would be bad for liver and kidney of mice.So Potassium Oxonate 250 mg/kg(ip.) was used for modeling acute hyperuricemia.The level of BUA, CR, BUN in Xanthine+Potassium Oxonate group higher than normal group obviously.Xanthine 600mg/kg(ig.)+ Potassium Oxonate lOOmg/kg (sc.) is the best way to induce chronic hyperuricemia.2. TCCF can obviously reduce the level of BUA in acute hyperuricemia mice and 220 mg/kg is the best dosage.TCCF can inhibit the activity of XOD and 220 mg/kg is the best dosage.TCCF has no effct on ADH and GDH.3. TCCF can obviously reduce the level of BUA in chronic hyperuricemia mice and 440 mg/kg is the best dosage. TCCF can obviously reduce the level of UUA in chronic hyperuricemia mice and 220 mg/kg is the best dosage. TCCF can inhibit the activity of XOD and 440 mg/kg is the best dosage.The level of CR and BUN in TCCF(440 mg/kg and 220 mg/kg) group was a little lower than model group, but the result has no statistics sense.Conclusion:1. Potassium Oxonate 250 mg/kg(ip.) can establish the acute hyperuricemia of mice with much effective and safe.2. TCCF can obviously reduce the level of BUA in acute and chronic hyperuricemia mice.The mechanism may be corrclated with inhibiting the activity of XOD.3. TCCF would not increase the damage of kidney.TCCF was shown to have protective and preventive effect on uric acid nephropathy.
