Purpose:1. In fifty-five Lactobacillus stains from department of micro-ecologyof Dalian Medical University, we screened for strain with higher decompositionnucleoside rate. Through the biological characteristics selected, DM9218is theoptimal strain.2. Observe the influence of DM9218to a rat model of hyperuricemia.3. Study the preliminary safety of DM9218.Method:1. Screened for strain with higher decomposition nucleoside rate andthe biological characteristics selected. Based on RP-HPLC method, by detecting themedium of inosine, guanosine content changes before and after in an hour, screeningwhich has the fastest nucleoside decomposition rate of Lactobacillus strains as acandidate strain.The biological characteristics include acid-resistant test, choline-resistant test,inhibitory test of target strain fermentation liquid on pathogenic bacteria, intestinalenzymes (trypsin, and pepsin)-resistant, antibiotic sensitivity test, HT-29colon cancercell adhesion assay and H2O2determination test.2. The influence of optimal strain to a rat model of HyperuricemiaThe cleaning Wistar rats were randomly divided into five groups: normalcontrol group, Hyperuricemia model group, Lactobacillus with low dose interventiongroup, Lactobacillus with high dose intervention group, Allopurinol interventiongroup. As building Hyperuricemia model as rat gastric perfusion in Lactobacillussuspension. Draw blood from the vein in experiment0d,7d,14d.Determination ofplasma uric acid (UA), blood urea nitrogen (BUN), creatinine (Cr) content.3. Study the preliminary safety of optimal strain.Study the preliminary safety of optimal strain includes three parts: acute toxicitytest, bacterial translocation test and the detection of the plasmid. 4. Identification of optimal strainCombine16SrDNA sequencing with biochemical reaction characteristics ofsugar.Determine the type of optimal strain.Results:1. From55strains of Lactobacillus strains were screened for4strainswith higher nucleotide decomposition rate strains: DM9218, DM9242, DM9505, andDM9514.As result of the biological characteristics in four candidate strain, DM9218hasthe largest nucleoside decomposition rate, domesticated tolerance of gastrointestinaltract environment, certain antibacterial ability, multiple antibiotics sensitivity, andgood in cell adhesion. So, DM9218is the optimal strain.2. The influence of optimal strain to a rat model of hyperuricemia.The contentof serum uric acid of Lactobacillus with low dose intervention group is obviouslylower than that of Hyperuricemia model group(P<0.05),but obviously higher thanthat of Allopurinol intervention group(P<0.05).The content of serum uric acid ofLactobacillus with high dose intervention group is obviously lower than that ofHyperuricemia model group(P<0.05),also lower than that of Lactobacillus with lowdose intervention group, but obviously higher than that of Allopurinol interventiongroup(P<0.05).There are no significant differences in both blood urea nitrogen(BUN) and serum creatinine (Cr) content(P>0.05).3. In preliminary safety evaluation experiments, DM9218was not found inbacterial translocation. In virulence experiments, mice were normal activities, notoxicity or mortality occurred. Plasmids were not found in the strain DM9218.4.16SrDNA sequencing shows that DM9218is Lactobacillus plantarum.Conclusion:Directed screening to55strains of Lactobacillus,DM9218has thelargest nucleoside decomposition rate, domesticated tolerance of gastrointestinal tractenvironment, certain antibacterial ability, multiple antibiotics sensitivity, and good incell adhesion.Also,DM9218was not found in bacterial translocation. In virulenceexperiments, mice were normal activities, no toxicity or mortality occurred. Plasmidswere not found in the strain.Animal experiments showed that: the DM9218strainswith a certain role in the prevention and treatment of hyperuricemia Wistar rats.DM9218is Lactobacillus plantarum.
