Object: To detect the Notch4 methylation status of peripheral blood in Uygur patients with Pigeon breeder's lung and to explore its clinical significance.Methods: The data and blood samples were enrolled as described in Kashi Prefecture from 2015 to 2016.?60 subjects including 20 cases of Uygur Pigeon breeder's lung patients were diagnosed as the case group,20 cases of healthy persons in Uygur feeding pigeons and 20 cases of healthy persons in Uygur who not raise pigeons as control groups?.And all the blood samples were extracted,conventional DNA extraction,bisulfite conversion,PCR amplification,transcription in vitro and RNase a specific enzyme cut,parallel matrix assisted laser desorption ionization time of flight mass spectrometry mass spectrometry technique was used to detect Notch4 gene methylation status analysis.Results: Detection of Notch4 fragment of CpG loci in total 6?CpG1,CpG2,CpG3,CpG4,CpG5,CpG6?.CPG1 ? CpG4 ? CpG5 ?CpG6 in three groups of samples methylation distribution difference was not statistically significant?P > 0.05?.CpG2/3 in three samples of methylation distribution difference was statistically significant?p<0.05?.The methylation rate of Cp G2 /3 of Uygur feeding pigeons lung patients higher than the Uygur feeding pigeons healthy personsis,the difference has statistically significant?P <0.05?.The methylation rate of Cp G2 /3 of the Uygur feeding pigeons lung patients higher than the healthy group,the difference has statistically significant?P <0.05?.There is no difference in the methylation rate of CpG2/3 between the Uygur feeding pigeons healthy personsis and the healthy group,the difference has no statistically significant?P >0.05?? Conclusion: Notch4 gene methylation by inflammation and signaling pathways that control the occurrence of lung fiber development.
