Objectives To investigate the association of the nucleotide polymorphisms (SNPs) at the genes forinterleukin-10(IL-10) and IL-2with the variable susceptibility to Uygur patients with pigeon breeder’s lung.Methods The data were collected as described (The total included92cases, of which62casesfeeding the pigeons,30cases of the normal control group), DNA was isolated from blood samples.Primerdesign, sample preparation, PCR, SAP and single base extension were performed by using MassARRAYtechnology according to SNP loci appendix sequence information,SNPs at the genes for IL-2(rs2069762)and IL-10(rs1800896)were analyzed using mass spectrometry.Statistical analyses were performed by withSPSS17.0statistical software, genotypes and allele frequencies at each SNP were compared by using thechi-square test, and calculated the relative risk value(OR). Results subjects with the GG GA AAgenotype at IL-10-1082G/A were found which were tested for Hardy-Weinberg equilibrium (HWE).Thosesubjects with the GG GT TT genotype at IL-2-330G/T.2.There were no significant differences in subjectwith GG GA AA genotype and the frequencies of genotypes at each SNP at rs1800896among threegroups(χ2=6.346,P=0.175;χ2=4.685,P=0.096), subjects with GG GT TT genotype at rs2069762showeda significant difference in the distribution of genotype among three groups(χ2=14.369,P=0.006),nodifference in the distribution of the alleles of the G、A among groups(χ2=0.061,P=0.97). Subjects with GGGT TT genotype at rs2069762showed a significant difference in the distribution of genotype between PBLand pigeon breeders groups or healthy control (χ2=13.413,P=0.001;χ2=8.783,P=0.012),while nodifferences between pigeon breeders group and healthy control (χ2=0.619,P=0.734).4. the relative risks(RR) of PBL in case group compared to the pigeon breeder’s group with the heterozygotes GThomozygous GG,TT genotype occurred relative risk OR (95%CI) were1.567(1.082-2.295),0.309(0.127-0.755),P=0.002,0.005, and the difference was statistically significant; homozygous GG and TT differencewas not statistically significant; We observed that the relative risks of PBL in case group compared withcontrolled group with the heterozygotes GT to GG genotype was1.154(P=0.004),95%CI:1.047-2.268,which indicated a significant difference. However, Heterozygous and homozygous TT/GT was nosignificant difference (P=0.671),No significant meaning as well as between homozygous GG/TTdifference. Conclusion1. IL-10(rs1800896)、IL-2(rs2069762) gene polymorphism in the presence ofUighur population.2. not find the presence of IL-10(rs1800896) gene polymorphisms and susceptibilitybetween PBL, need further study.3.IL-2gene polymorphisms and susceptibility between PBL, mayinfluence the level of IL-2gene transcription changes in PBL play a role in the pathogenesis.4. IL-2genepolymorphisms found in: GT heterozygous genotype may be a risk factor predisposing PBL, carryingheterozygous GT may increase the risk of suffering from PBL,and the need to increase further validationsample size...
