Objective:In this study,we investigated the expression of mt-P53,Mcm7 and Aurora-A genes in normal colorectal mucosa,colorectal adenoma and colorectal adenocarcinoma,and analyzed the relationship between the expression and the clinicopathologic features of colorectal adenocarcinoma.We evaluated the effect of mt-P53,Mcm7 and Auroa-A on carcinogenesis,progression and prognosis in colorectal adenocarcinoma.Methods:The mt-P53,Mcm7 and Aurora-A gene were detected by using immunohistochemical S-P methods in colorectal adenocarcinoma tissue,colorectal adenoma and normal colorectal tissue.And we analysed the relationship between their expression and prognosis in colorectal adenocarcinoma.Results:1.Ectopic expression rates of mt-P53,Mcm7 and Aurora-A genes in normal colorectal mucosa,adenomas and colorectal adenocarcinoma rose in turn(P<0.05).The positive expression rates of of mt-P53,Mcm7 and Aurora-A in each colorectal lesions groups(a low level,high level of adenoma,well-differentiated adenocarcinomas,moderately differentiated adenocarcinoma,poorly differentiated adenocarcinoma)gradually increased(P<0.05).2.The expression of P53 genes in colorectal adenocarcinoma tissues to be associated with the depth,Dukes stages and lymph node metastasis(P<0.05),mt-P53 expression in patients with less than five-year survival curve was higher than those with more than or equal five-year survival curve(P<0.05).But there was no correlation with the patients' age,gender,size of tumors and differentiation degree(P>0.05).The expression of Mcm7 genes was associated with the depth,differentiation degree,Dukes stages and lymph node metastasis in colorectal adenocarcinoma tissues(P<0.05),but there was no correlation with the patients' age,gender and size of tumors(P>0.05).The positive expression of Mcm7 in patients with less than five-year survival curve and those with more than or equal five-year survival curve was no significant differcece(P>0.05).The expression of Aurora-A gene was associated with the depth in colorectal adenocarcinoma tissues(P<0.05),and the positive expression in patients with less than five-year survival curve was higher than those with more than or equal five-year survival curve.The difference had statistical significance(P<0.05).But there was no correlation with the patients' age,gender,size of tumors,differentiation degree,Dukes stages and lymph node metastasis(P>0.05).3.The positive expression rate of Mcm7 in high differentiated colorectal adenocarcinoma tissues was significantly higher than that in high level of colorectal adenoma.The difference was statistically significant(P<0.05).The positive expression of mt-P53 and Aurora-A in high differentiatied colorectal adenocarcinoma and high level of colorectal adenoma were no significant difference(P > 0.05).4.In colorectal adenocarcinoma,the expression of Mcm7 and Aurora-A genes were more significantly related to the expression of mt-P53 gene(r=0.284,P<0.05;r=0.547,P<0.01),but the expression of Mcm7 gene had no significant relation with the expression of Aurora-A gene(P>0.05).Conclusion:1.The positive expression rate of mt-P53 and Aurora-A in colorectal adenocarcinoma is significantly higher.It suggests that mt-P53 and Aurora-A may be closely related to the occurrence of colorectal adenocarcinoma,and they may become a useful target for prognostic in colorectal adenocarcinoma.2.The abnormal expression of Mcm7 protein in colorectal adenoma and colorectal adenocarcinoma prompts Mcm7 can be used as an evaluation index of colorectal malignant degree,and is conducive to identify the high differentiated colorectal adenocarcinoma and high level colorectal adenoma.3.In colorectal adenocarcinoma,the expression of mt-P53 and the expression Mcm7 and Aurora-A have positive correlation.It prompts that the correlated mechanism of mt-P53 and Mcm7,Aurora-A plays a role in colorectal adenocarcinoma.Combined detection of the three proteins may be a reference index that judges biological behaviour of colorectal adenocarcinoma and determines the clinical treatment plan.
