Objective1,To observe the expression of Yes-associated protein (YAP) in the normal colorectal tissue,adjacent nontumor tissue, colorectal polyp, colorectal adenoma and the colorectaladenocarcinoma. and to understand its significance in tumors development procession.2,To investigate the apoptosis in the colorectal adenocarcinoma, and to understand its mechanismand significance.3,To observe the expression of Yes-associated protein (YAP), its relationship with theproliferation and apoptosis of tumor cells and its clinical significance in colorectaladenocarcinoma.MethodsThe expression of YAP in 10 normal colorectal tissue, 10 adjacent nontumor tissue, 20colorectal polyp, 40 colorectal adenoma and 80 colorectal adenocarcinoma were detected byimmunohistochemical technique, and apoptosis was observed by terminaldeoxynucleotidyltransferase-mediated dUTP nick-end labeling ( TUNEL ) in the 80 colorectaladenocarcinoma tissues.Results1,The positive expression rate of YAP was 0.0% in normal colorectal tissues, 0.0% in adjacentnontumor tissues, and 62.5% in colorectal adenocarcinoma. And the positive expression rate ofYAP in 20 colorectal polyp, 40 colorectal adenoma was 5.0% and 32.5%,respectively. Therewere significant differences among them ( P < 0.05). The expression rate of YAP in the olorectaladenocarcinoma was the highest than other tissues. The positive expression rate of YAP innormal colorectal tissues, adjacent nontumor tissues and colorectal polyp tissues were nosignificant differences, but they were all lower than the colorectal adenoma.2,In the colorectal adenoma group, the expression of YAP in the mild, moderate and severeatypical hyperplasia were significant differences (P < 0.05). There was no differences ofoverexpression of YAP in colorectal adenocarcinoma with the tumor differentiation (P>0.05),however, the expression lever of YAP was higher from highdifferentiation to poor differentiation.there were no significant differences in patients'age, sex and tumor size (P >0.05).3,The morphology of colorectal adenocarcinoma cell showed changes such as nuclearchromatin condensation and fragmentation by the TUNEL. The apoptosis rate was ( 0.85±0.23 )% in colorectal adenocarcinoma. In the colorectal adenocarcinoma, the apoptosis rat of highdifferentiation tissues was (1.03±0.18)%, the moderate differentiation tissues was (0.84± 0.21)%, the poorly differentiation tissues was (0.65±0.09)% . Along with the differentiationfrom high to poorly, the apoptosis rat was also correspondence from high to low.4,There were significant decline in apoptosis rate in colorectal adenocarcinoma with positiveexpression of YAP (P < 0.05).ConclusioConclusion1,YAP plays an important role in the inhibition for the cell apoptosis and occurrence andprogression of human colorectal adenocarcinoma, and is useful for early diagnosis target.2,YAP also is the diagnosis target for the adenocarcinoma differentiation and the adenoma withdifferent hyperplasia.3,Along with the adenocarcinoma differentiation, the apoptosis rat was also correspondence fromhigh to low.
