ObjectiveThe expressions of EGFR,Lyn and MCM7 were detected in 196 cases of the colorectal cancer tissues and the matched normal tissues adjacent to cancer tissues.We analyzed the different expressions of EGFR,Lyn and MCM7 between two groups of tissues and the relationship between the expressions in the colorectal cancer tissues and the clinicopathological parameters of the patients with colorectal cancer.The predictive significance of the expressions of three molecules in the colorectal cancer tissues for the clinical prognosis was also be analyzed.This study intends to investigate the effective evaluation indexes for the prognosis of the colorectal cancer patients,which will make the colorectal cancer research more targeted in future.MethodsUsing the tissue chips prepared from collected specimens of the colorectal cancer tissues and the matched normal tissues adjacent to the cancer tissues,the expressions of EGFR,Lyn and MCM7 in the colorectal cancer tissues and the matched normal tissues adjacent to the cancer tissues were detected by the immunohistochemical method.The SPSS statistical software package was used to statistical analyze the differences and the relationships of the expression of EGFR,Lyn and MCM7 between the two groups of tissues,and also to analyze the relationships and significances of EGFR,Lyn and MCM7 expressions in the colorectal cancer tissues with clinicopathological characteristics,patients survival and prognosis.Results1.The expression levels of EGFR,Lyn and MCM7 were all significantly higher in the colorectal cancer tissues than that in the matched normal tissues adjacent to the colorectal cancer tissues,and there were significant differences in expression between the two tissues(P<0.001).2.The expressions of EGFR,Lyn,and MCM7 in the colorectal cancer tissues were significantly positively correlated with each other(?~2=6.144,P=0.046),and their expression levels were related to the tumor location(P<0.001),which of the left-sided colorectal cancer was significantly higher than the right-sided.3.The expression level of EGFR was related to the invasive depth of tumor(P=0.017),the lymph nodes metastasis(P<0.001)and the tumor TNM stage(P=0.033);the expression level of Lyn was related to the lymph nodes metastasis(P=0.026)and the tumor TNM stage(P=0.019);the expression level of MCM7 was related to the degree of tumor cell differentiation(P=0.038)and the tumor TNM stage(P=0.035).4.The Kaplan-Meier method was used to analyze the survival status of the patients,and the Log-rank test was performed.The results showed that low expression level of EGFR(P<0.001),Lyn(P=0.012)and MCM7(P=0.043)showed the better OS than the high level.5.The multivariate Cox proportional hazard regression analysis indicated that the expression levels of EGFR(HR=4.414,95%CI:2.136~9.052,P=0.026),Lyn(HR=1.622,95%CI:1.125~4.036,P=0.041)predicted for the death of the colorectal cancer patients;and the expression level of MCM7 was not the independent risk factor for the OS in the patients with colorectal cancer(HR=0.951,95%CI:0.492~1.84,P=0.882).Conclusion1.The expression levels of EGFR,Lyn and MCM7 in the colorectal cancer tissues were higher than those matched normal tissues adjacent to the colorectal cancer tissues,the expression levels of them in colorectal cancer tissues were positively corelated to each other.2.The expression levels of EGFR,Lyn and MCM7 in colorectal cancer tissues were related to the tumor TNM stage and the tumor location,which of the left-sided colorectal cancer were significantly higher than the right-sided.In addition,in the colorectal cancer tissues,the expression level of EGFR was related to the invasive depth of tumor and the lymph nodes metastasis,the expression level of Lyn was related to the lymph nodes metastasis,and the expression level of MCM7 was related to the degree of tumor cell differentiation.3.Survival analysis showed that the OS in the colorectal cancer patients with the lower expression levels of EGFR,Lyn and MCM7 in the colorectal cancer tissues was better than the higher.Multivariate proportional hazard regression analysis showed that the expression levels of EGFR and Lyn were independent risk factors for the OS in the patients with colorectal cancer and the expression level of MCM7 was not.
