| ObjectiveThis study established CUMS with solitary depression model rats,to observe the behavioral changes and the content of 5-HT in the hippocampus,serotonin and ACTH in serum and the changes of CORT content,the anti depression effect and mechanism of gentianella acuta possible.MethodsAll 40 Wistar female rats were randomly divided into five groups:blank control group,model group,high dose treatment group,low dose treatment group and positive drug group.Except the blank group,each rat was solitary and accept the stimulation of the stressor,including:24hours water deprivation,24 hours of fasting,40 ? high temperature 5min,swimming in 4? ice water for 5min,electric shock for 5min,day and night upside down,24h wet pad for a total of 7 different stimuli in 21 days 7 kind of stimulation were given daily,given 1 kinds of stimuli,each stimulus can occur up to 3 times,with a continuous stimulation,to prevent the animal can expect a stimulus to happen.From the beginning of the eighth day,a drug was given once a day.The high dose group was given 3g/kg gentianella acuta and the low dose group was given1.5g/kg gentianella acuta.The positive group was given fluoxetine hydrochloride 1.8mg/kg.After 21 days,the changes of body weight were observed before and after the formation of the rats in each group,and the bilateral hippocampus was decapitated and the content of 5-HT was measured.Orbital rapid blood and determinate the content of ACTH and CORT.Results1.Behavior:twenty-one days later,the rats in the model group weight decreased;rats still forced swimming test exercise time increased,the degree of preference for sugar decreased;distance in the central region the sum becomes small.The weight of the rats in each treatment group was higher than that of the control group,and the preference for sugar syrup was increased.The time of inactivity was decreased in the forced swimming test.2.Weight change:there was no significant difference in body weight between the groups(P>0.05).During the experimental period,compared with the blank group,the weight of the model group decreased,there was significant difference(P<0.05).Compared with the model group,there was no significant difference in the weight of the rats in the other groups.3.Preference of sugar syrup:before the experiment,there was no significant difference between the two groups(P>0.05).After the experiment compared with the control group,model group,sucrose preference rats decreased significantly(P<0.01);compared with the model group,gentianella acuta high and low dose group of sucrose preference rats were significantly increased(P<0.05).4.The content of 5-HT:the concentration of 5-HT in hippocampus of rats in each group was increased.5.In the serum of ACTH:21 days,compared with the model group,the content of ACTH in serum of rats in high dose group decreased significantly(P<0.01);the content of serum ACTH was significantly decreased in low dose group(P<0.05).6.In the serum of CORT:21 days,compared with the model group,the content of CORT in serum of rats in high dose group decreased significantly,there was significant difference(P<0.01);the content of serum CORT decreased in low dose group,there was significant difference(P<0.05).Conclusion1.To establish a model of CUMS combined with solitary depression in rats.The anti depression effect of the extract on the depression model rats was the best in the high dose group.2.Reduce the content of 5-HT in rat brain,indicating the onset of depression and five hydroxytryptamine content change is closely related to,and consistent with the hypothesis of 5-HT;reduce the serum ACTH and CORT content,indicating gentianella acuta improves the function of HPA axis,HPA axis reduces high activity,inhibition of excited state. |
