Objective:To observe the influence of chronic alcoholic liver damaged rat model byGentianella Acuta compatibility of Semen Hoveniae and to explore its action mechanism.Methods:1.Through Gentianella Acuta compatibility of Semen Hoveniae (1:1,2:1 and 1:2) respectively on chronic alcoholic liver damaged rat model, to observe the characteristics of a rat liver pathological physiological changes and the effect of serum ALT, AST values change, and the serum TG, CHO’s changes,to find the better compatibility combination which has better therapeutic effect on chronic alcoholic liver damaged rat model.2.To observe Gentianella Acuta compatibility of Semen Hoveniae’s influence of rat serum IL-6 and TNF-a content,CYP2E1 protein expression and mRNA expression, and discuss the Gentianella Acuta and Semen Hoveniae compatibility mechanism on chronic alcoholic liver damage.Results:1. Gentianella Acuta compatibility of Semen Hoveniae could improve a chronic alcoholic liver damaged rat model’s pathological state,1:1 group had the most obvious effect.2. The prescription of Gentianella Acuta compatibility of Semen Hoveniae can reduce serum ALT and AST values of chronic alcoholic liver damaged rat model,1:1 group had the most obvious effect.3. The prescription could reduce chronic alcoholic liver damaged rat model serum TG, CHO values.1:1 group reduced TG value had the best effect,2:1 group reduced Cho value effect was the best. It was illustrated the good effect on liver lipid metabolism in rats, and its mechanism may be related to the inhibition of lipid peroxidation.4. In the study of the mechanism, Gentianella Acuta compati-bility of Semen Hoveniae rescription could reduce chronic alcoholic liver damaged rat model serum IL-6 and TNF-αvalue, indicating that it could help regulate the disorder of cytokines, restore the body to promote the balance of inflammatory cytokines and prevent the apoptosis and necrosis of liver cells. The 1:1 group had the best effect on reducing the IL-6 value, and the 2:1 group had the best effect on reducing the TNF-α.5. In the study of the mechanism, the prescription of Gentianella Acuta compatibility of Semen Hoveniae 1:1 group could significantly down regulated in chronic alcoholic liver damaged rat model for CYP2E1 protein expression and mRNA expression.Conclusion:1. Gentianella Acuta compatibility of Semen Hoveniaeprescription could improve ALD model in rat liver tissue pathological state and regulatetheALT,AST,TG,CHO,IL-6,TNF-alevel which was in serum, indicating that it has good therapeutic effect on ALD.2 Theoptimum proportion of Gentianella acuta and Semen Hoveniae prescriptionis 1:1.3. TheGentianella Acuta and Semen Hoveniae compatibility group of proportion 1:1 can inhibit the activity of CYP2E1 and its mechanism may be related to anti lipid peroxidation.
