ObjectiveThe study was based on the establishment of CUMS combined with the model of depression in rats.To explore the way and mechanism of anti depressant gentianella acuta and Acorus tatarinowii compatibility by the method of experimental techniques in immunology and molecular biology.To provide experimental basis for the research and development of clinical antidepressant Chinese medicine.MethodDetermine the optimal ratio of Gentianella acuta and Acorus tatarinowii compatibility from the rat behavior and the contents of monoamine neurotransmitters. Observe the neuronal morphological structure of Rat hippocampus and mPFC by light microscope and Electron microscope. Determine the expression of Neuronal apoptosis and Neurotrophic factor by the method of Immunohistochemical. Determine the expression of Hippocampal p-CREB, BDNF and TrkBmRNA by RT-PCR.Result1.Behavior:Modeling Chronic stress of model group for 21 days,rats loss their weight;The sucrose preference degree decreased;Forced swimming time extended;The total distance of the central activity became shorter.The 1:11:2 2:1 ratio group of Gentianella acuta and Acorus tatarinowii,there was increasing trend of CUMS combined with solitary rat;The sucrose preference degree increased;The total distance of the central activity became longer.2.Contents of monoamine neurotransmitters.The 5-HT and DA content of Rat hippocampus and mPFC group had varying degrees of improvement,The 1:1 and 1:2 group had a increasing of GAB A content and a reducing of Glu content.1:2 group effect is the best.3.Pathological changes.Under light microscope:The Rat hippocampus and mPFC of HE staining model group had Permutation of disorder, increasing, karyopy-knosis.The apoptosis of subcell for compatibility group reduced significantly.Compared with model group,the Rat hippocampus and mPFC of TUNEL staining model group increased the numbers of apoptosis neurons significant.Compared with model group, the group was significantly decreased.Under electron microscope:The inner and outer membrane damaged of hippocampal and mPFC cells in model group mitochondrial inner and outer membranes or fusion,ridge fracture or swelling;Synaptic fusion, disappear-rance of the gap, or reduction of synaptic vesicles or nerve felt edema.Compatibility group had clear cell membrane;Mitochondrial bilayer membrane structure was clear,normal ridge structure;Synaptic dense substance, the existence of synaptic space.4.Immunohistochemical:Experimental results of immunohistochemistry show,the protein expression of Caspase-3 and Bax in hippocampus and mPFC of compatibility group was significantly lower than that in model group,the protein expression of Bcl-2, BDNF and p-CREB was significantly higher than that of model group.5.RT-PCR:The levels of TrkBmRNA, CREB and BDNF in hippocampus and mPFC in compatibility group were higher than those in model group.Conclusion1.The rat model of depression was successfully established.The antidepressant effects of Gentianella acuta and Acorus tatarinowii compatibility on depression model rats which best is 1:2 group.2.Gentianella acuta and Acorus tatarinowii compatibility could effectively protect and reversal in the rat hippocampus and mPFC neurons damage, prevent and inhibit neuronal apoptosis, its mechanism may be related to the protection of neurons and neural plasticity.3.Gentianella acuta and Acorus tatarinowii compatibility group antidepressant effects may be related to the activation of BDNF/TrkB mediated signaling pathway.
