Objective:The study was based on the Gentianella acuta,through the Gentianella acuta pharmacodynamics experiments of type of adjuvant arthritis model in rats.Methods:The rats is a model of an Adjuvant Arthritis model,SD rats were randomly divided into model control group,blank model group,positive control group(multi-glycosides of tripterygium wilfordii),the Gentianella acuta high dose group,the Gentianella acuta middle dose group,Gentianella acuta low dose group,each group of eight.In addition to outside the rest of the blank group rats right rear fully plantar subcutaneous injection freund's complete adjuvant(CFA)0.1 ml inflammatory,copying the AA models,blank group injected with saline.From 14 days began to fill the stomach,gastric 18 consecutive days.Regularly measuring the rat body weight,fully right toe swelling degree,ankle and spleen pathological HE,Elisa method to detect the rats in the serum TNF-?,IL-10 levels,Immunohistochemical method to detect of VEGF in synovial tissue,ICAM-1 and MMP3 protein expression.Result:1.Gentianella acuta each dose group can increase the weight of AA rats,reduce AA rats toes swelling,lower grade AA rats arthritis,reducing the generation of AA rats spleen white pulp.2.Gentianella acuta each dose group can reduce the content of AA rats in the serum TNF-a and improve the content of serum IL-10 of each dose group of AA rats to observe the level of cytokine.The high dose group of the most obvious effect.(P<0.05)3.Gentianella acuta each dose group can reduce the AA rats synovial tissue VEGF,ICAM-1,MMP-3 protein expression,so as to achieve lower synovial inflammation,reduce joint swelling and delay the purpose of the joint bone destruction.The high dose group of the most obvious effect.(P<0.05,P<0.01)Conclusion:Gentianella acuta can effectively restrain secondary joint swelling,can significantly reduce the content of AA rats in the serum TNF-? and improve the content of serum IL-10 in synovial tissue,VEGF,MMP3 ICAM-1 protein expression regulation function to alleviate synovial inflammation,relieve joint swelling,joint and delay the basic pathology of bone destruction,together leaves off gentian high dose group is better than that of middle dose group and low dose group.
