Objective:To observed an anti-inflammatory effect of the Gentianella acuta (Michx.) Hulten to myocardial ischemia-reperfusion injury model rats, we explored its active mechanism.Methods:1.We used the high, medium and low dose group ethanol extract of the Gentianella acuta (Michx.) Hulten respectively to the myocardial ischemia-reperfusion injury model rats, and observed the effect of CK-MB, cTNI and myocardial infarction area in the serum of rats to screen out the anti-inflammatory effective dose of the Gentianella acuta (Michx.) Hulten.2.We observed the effect of the myocardial pathological morphology and the ultra micro struct of myocardial by applied the anti-inflammatory effective dose of the Gentianella acuta (Michx.) Hulten to the myocardial ischemia-reperfusion injury model rats, getting a conclusion that the Gentianella acuta (Michx.) Hulten had a good protection to the myocardial ischemia-reperfusion injury model rats.3.By used the Western Blot, immunohistochemical, RT-PCR, we detected the effect of the distribution and expression of the protein, ether the expression of mRNA, to NF-κB, COX-2 in the myocardium with the anti-inflammatory effective dose of the Gentianella acuta (Michx.) Hulten. And then we could discuss the active mechanism to the myocardial ischemia-reperfusion injury model rats of the effective dose of the Gentianella acuta (Michx.) Hulten.4.By observed the effect of the effective dose of the Gentianella acuta (Michx.) Hulten to the TNF-α, IL-1β, IL-6, IL-10 contents in the serum of the myocardial ischemia-reperfusion injury model rats, we explored its active mechanism.Results:1.Compared the high dose, middle dose group with the model group, the effective dose of the Gentianella acuta (Michx.) Hulten could reduce the contents of the CK-MB、cTnl in the serum of the myocardial ischemia-reperfusion injury model rats significantly all, also could reduce myocardial infarction area.2.Compared with the high dose group and the model group, the high dose group could improve myocardial pathology morphology and ultra micro struct of myocardial of the myocardial ischemia-reperfusion injury model rats observably.3. Compared with the high dose group and the model group, the high dose group could induce the contents of the NF-κB, COX-2 in the myocardium of the myocardial ischemia-reperfusion injury model rats which distributed the protein also expressed the protein and mRNA observably.4.The high dose group of the Gentianella acuta (Michx.) Hulten could induce the contents of the TNF-α, IL-1β, IL-6, and also increase the contents of the IL-10 in the myocardium of the myocardial ischemia-reperfusion injury model rats observably.Conclusions:1.The high dose group had the most important, that explained the high dose group being the most effective dose group.2.The high dose group could improve myocardial pathology morphology and ultra micro struct of myocardial of the myocardial ischemia-reperfusion injury model rats observably, that explained it had a good protection to the myocardium of the myocardial ischemia-reperfusion injury model rats.3. Gentianella acuta (Michx.) Hulten could induce the contents of the TNF-α, IL-1β, IL-6, and also increase the contents of the IL-10 in the myocardium of the myocardial ischemia-reperfusion injury model rats observably, that explained its active mechanism could be inhibiting signal transduction pathway of NF-κB/COX-2, then cut the expression of inflammatory factors of transcriptional regulation.
