Cervical cancer is a major threat to women's life and health,and it is the fourth fatal cancer of females.Cervical cancer is difficult to detect because its symptoms are not distinct.Unfortunately,once symptoms have been detected,it is a terminal cancer.The incidence of cervical cancer is often accompanied by high-risk human papillomavirus(HPV)infection,which vulnerable to human epidermis and mucosal cells.The low-risk HPV can cause genital warts,high-risk HPV can cause cervical cancer.High-risk HPV 16 and 18 are the most common.Chinese medicine compound preparation which has a good development prospects because of its small side effects.The E6 gene of HPV 18 contained in Hela cells was constructed by bioinformatical tools.The molecular dynamics simulations of HPV 18 E6 and HPV 16 E6 were carried out.It was found that the key LOOP were acting as the role of "gatekeeper".The controller of small molecules accessing to protein binding sites for the treatment of HPV infection,which provides a theoretical basis for therapies.Natural Plant Antimicrobial solution(PAMs)is mainly made of Carthamus tinctorius,Lithospermum erythrorhizon,Blumea balsamifera,Solarium indicum and other medicinal plants of ethanol extract.As a consequence of its complex composition and wide adaptability,it is not clear about the specific efficacy of ingredients.In this paper,according to the effect of natural plant antibacterial solution on anti-tumor and skin repair and using high performance liquid chromatography(HPLC)to detect the source of medicinal materials,the content of hydroxy-safflower yellow A and aconitine was 33.87±1.01?g/ml and 22.06±0.06?g/ml respectively,and can not detect the bulleyaconitine?shikonin?loureirin?diosgenin in the sample.Secondly,the inhibitory effect of PAMs K3 on the phage was determined by M13 as the virus model.The results showed that,PAMs K3 had a strong inhibitory effect on phage,whose inhibition rate was 72.38%at 5%concentration.Hela cells were used as the research object to study the inhibitory effect of PAMs on tumor cells.Morphological observation and CCK-8 method showed that PAMs had a strong inhibitory effect on proliferation of tumor cells.Compared with the blank group,there was a significant difference from the control group,and the inhibition rate was 74.25%(48 h at 5%).The results of wound scratch assay and Transwell chamber migration showed that the natural plant antibacterial solution had a good inhibitory effect on the migration of tumor cells in vitro and has statistically significant difference,and the inhibition rate was 65.9%.Finally,the role of PAMs was studied by constructing a model of transplanted Hela cells in nude mice.The results showed that PAMs could slow down the tumor growth rate,and the organ index showed no significant difference compared with the model group.Moreover,PAMs could down-regulate the serum VEGF and inhibit the tumor growth.Its further molecular mechanism of action need to be further studied.
