Objective:Amomum villosum leaf oil was volatile oil extracted from leaves of Amomum villosum Lour.from Zingiberaceae by Steam distillation.It was colorless or light yellow clear liquid,Spicy and smelled like Fructus Amomi.It was used to promote digestion,eliminate abdominal distention,stop vomiting and so on.The present studies of Amomum villosummoanly concentrated in Fructus Amomi,so there were few studies on chemical components and pharmacological activities of Amomum villosum leaf oil.In this study,chemical components and pharmacological activities of Amomum villosum leaf oil were researched,which could provide theoretical basis for comprehensive development and utilization of Amomum villosum leaf oil and improving the utilization rate of A.villosum resources.Method:In the present study,Gas Chromatograph-Mass Spectrometer method was used to analyse chemical components and relative contents of Amomum villosum leaf oil.The DPPH radical scavenging activity,reducing power and total antioxidant capacity were measured by UV spectrophotometry to evaluate antioxidant activity in vitro of Amomum villosum leaf oil.Established mice ear swelling model induced by xylene and calculated the average ear swelling degree and ear swelling inhibition rate,investigated the anti-inflammatory activity of Amomum villosum leaf oil.Established mice pain model induced by acetic acid solution and observed the number of each mouse writhing in 20 minutes,to investigate the analgesic effect of Amomum villosum leaf oil.Used nutritional semisolid paste as the gastrointestinal transporting marker,weighed the stomach to calculate the rate of the residue in stomach;then measured the length of the small intestine and the distance from the pylorus to the semisolid paste to calculate the small intestinal propulsion rate,to investigate the effects of Amomum villosum leaf oil on mice gastric emptying and small intestine propulsion function.The wounds on the back skin of rabbits were prepared,and the wound healing rate was measured and calculated on the 3rd day,the 7th day and the 13th day after operation.Take the blood from the ear vein on the 3rd day and the 13th day,ELISA method was used to detect the levels of serum TNF-a and TGF-β1.After wound healing the wound specimens were collected for HE staining,to investigate the effects of Amomum villosum leaf oil on rabbit skin wound healing.Result:1.Analysis results of Amomum villosum leaf oil showed that the main components of Amomum villosum leaf oil were alcohols,alkene and carbonyl compounds.From Amomum villosum leaf oil isolated 45 compounds,the higher content were Pseudo limonene,3-Carene,d-Limonene and myrtenol,their contents were 41.12%,24.17%,3.25%and 3.18%2.In vitro antioxidant results showed that Amomum villosum leaf oil has certain antioxidant activity,and the antioxidant capacity had obvious dose effect relationship.Compared with the vitamin C(Vc),the DPPH radical scavenging ability and reducing ability of Amomum villosum leaf oil were different.But the results of determination of total antioxidant capacity showed that the total antioxidant capacity of Amomum villosum leaf oil was similar to Vc.3.The results of pharmacological activity studies showed that Amomum villosum leaf oil and dexamethasone could significantly reduce xylene induced mouse ear swelling,indicated that Amomum villosum leaf oil had good anti-inflammatory effect.Amomum villosum leaf oil and indomethacin could significantly reduce the number of mouse writhing induced by acetic acid,indicated that Amomum villosum leaf oil had significant analgesic effect.Amomum villosum leaf oil has two-way adjustment function on the mice gastric emptying,the effect enhanced with the increase of the dose within a certain scope,but when the dose increased to a certain degree,it had no promoting effect on gastric emptying.Amomum villosum leaf oil had the promoting function of small intestine propulsion,the effect had obvious concentration-response relationship and was similar to Mosapride.Amomum villosum leaf oil could significantly shorten the rabbit wound healing time and promoted wound healing,its effect and was equal to that of nitrofurazone.Tissue section HE dyeing observation showed that amomum villosum leaf oil could promote wound healing,the newborn skin in good condition,with less inflammatory cells and tissue lesions.The determination results of rabbit serum TNF-α and TGF-β 1 showed that,the effect of Amomum villosum leaf oil on serum TNF-α and TGF-β 1 was similar to Nitrofurazone.Amomum villosum leaf oil could increase the content of serum TNF-α and TGF-β 1 in rabbits,up regulation of serum TNF-α and TGF-β 1 may be a mechanism for Amomum villosum leaf oil to promote wound healing.Conclusion:The researches showed that,the main components of Amomum villosum leaf oil were Pseudo limonene,3-Carene,d-Limonene and myrtenol.Amomum villosum leaf oil had certain antioxidant activity,anti-inflammatory and analgesic effects,and could promote gastrointestinal motility and wound healing.These showed that Amomum villosum leaf oil had good biological activity and worthy of further research.These researches would laid the theoretical foundation for the comprehensive development and utilization of Amomum villosum leaf oil. |
