| The Synsepalum dulcificum leaf can improve heartburn, indigestion, poorappetite and other symptoms, such as atherosclerosis, high blood pressure anddiabetes. It also can enhance liver function and immunity, which has obvious curativeeffect. In this paper, extraction, purification, antifatigue, antioxidant and antitumoralActivities of total flavones of Synsepalum dulcificum leaf from Hainan, China wasresearched. This study results could provide a certain theoretical foundation forcomprehensive utilization and further development of Synsepalum dulcificum leaf.The results of the studies as following:1. Nutritional compositions of Synsepalum dulcificum leaf was analyzed at thefirst time. The results showed that Synsepalum dulcificum leaf was rich in protein,fibre and polysaccharide. The total amino acid was8.65g/100g, and the essentialamino acid accounted for41.5%. This research indicated that Synsepalum dulcificumleaf was a kind of food or drug with rich nutrients and worthy of development andutilization.2. The chemical composition of Synsepalum dulcificum leaf essential oilobtained by steam distillation was determined and characterized the in vitroantibacterial activity and antitumoral activity of this essential oil. The major chemicalcompounds of leaf essential oil were spathulenol, limonene, diisooctyl phthalate,dibutyl phthalate, palmitic acid and linalool. The result of the antimicrobial assayshowed that the essential oil displayed varying degrees of antibacterial activity againstall tested bacterial except for Pseudomonas aeruginosa. In addition, the antitumoralactivity using MTT assay of the leaf essential oil exhibited this oil was effectiveagainst human K562cancer cell line in a dose dependent manner, its inhibitionconcentration50%(IC50) value was13.5μg/mL.3. The Synsepalum dulcificum leaf total flavonoid was determined and thetechnology and conditions was to be optimum by using the response surface methodon the basis of single factor experiment. A second order quadratic mathematicalmodel for Synsepalum dulcificum leaf total flavonoids were established and the best technical conditions were obtained as follows:ethanolic concentration57.09%, the ratio of liquid to solid30.73:1mL/g, ultrasonic time38.14min and extraction temperature61.40℃. The predicted polyphenol extraction yield was4.14%, under the optimized condition, the experimental yield was (4.03±0.03)%, which was in good agreement with its predicted counterpart.4. The purification of Synsepalum dulcificum leaf total flavonoids were established using the method of macroreticular resins. The major factors which had influence on dynamic adsorption and desorption of macroreticular resins were investigated. The results showed that HPD500was ideal as purification of Synsepalum dulcificum leaf total flavonoids, The optimal static adsorption and desorption conditions were obtained as follows:concentration of sample solution1.4mg/mL, feeding speed of sample solution1.5mL·-min-1, pH value of sample solution6.0, eluent feeding speed2.0mL·min-1, and eluent ethanol concentration80%. Unde the optimal conditions, the purification of Synsepalum dulcificum leaf total flavonoids as high as89.5%, and the yield was48.3%.5. The object of this paper was to investigate the effect of Synsepalum dulcificum leaf total flavonoids on fatigue in swimming-induced injury model in mice, and also characterize the in vitro antitumoral activity of total flavonoids. The results showed that Synsepalum dulcificum leaf total flavonoids can obviously prolong the time of swimming, especially high-dose group. In addition, Synsepalum dulcificum leaf total flavonoids can reduce the content of BUN, LD and MDA, enhance mice liver glycogen and the activity of CAT, SOD. the antitumoral activity using MTT assay of the leaf total flavonoids exhibited it was effective against human K562,SGC-7901, SPC-A-1cancer cell line in a dose dependent manner, its inhibition concentration50%(IC50) value was10.31ã€38.70and88.19μg/mL, respectively... |
