1 ObjectiveAmomi Fructus is dried ripe-fuit from Amomum villosum Lour., Amomum villosum Lour. var. xanthioides T.L. Wu et Senjen, and Amomum longiligulare T. L. Wu. It's well known as one of the four famous southern herbs in China. Amomum villosum Lour. is mainly produced in Guangdong, and the herbal produced in Yangchun is traditionally considered as the best in respect of quality. Amomum villosum Lour. var. xanthioides T.L. Wu et Senjen's main produced areas are Vietnam, Myanmar and Thailand. Amomum longiligulare T. L. Wu's main produced areas are Hainan and Guangdong. Because of low-yield and poor quality, both of the latter two kinds of Amomum villosume have hardly been demanded in the market.In recent years, Amomum villosum Lour. becomes the mainstream product in Chinese medicinal market.Amomum villosum Lour., a herb with warm property and pungent taste,has such effects as resolving dampness to improve appetite,warming spleen to arrest diarrhea,regulating Qi to prevent miscarriage etc.It has been used on clinic for more than 1300 years.In recent decades,domestic and foreign scholars did a lot of research on Amomi Fructus,but Amomum villosum Lour. from genuine producing regions lack of a complete standard of quality control. Therefore, in order to improve Amomum villosum Lour. medicine's standards to ensure its safe and reliable, stable and controllable, to ensure safe and effective on clinic, this article will make the three cultivated varieties of Amomum villosum Lour. which is produced in genuine regions, namely "changguo", "yuanguo" and "chunxuan" as a benchmark, compared to different origin and products of Amomum villosum Lour. medicine, evaluate Amomum villosum Lour. medicine's quality separately from the chemical quality evaluation (routine examination,chemical identification,determination,chemical composition analysis)and product specification quantitative method. Establishing chemical quality assesment standard of Amomum villosum Lour. from genuine producing regions and quantitative method of Amomum villosum Lour. product specification,provided a reliable basis for quality assesment of Amomum villosum Lour. medicine comprehensively.2 MethodsChemical quality evaluation:Refering to the approach of 2005 version of《The People's Republic of China Pharmacopoeia》, checking the water content,determining the essential oil content and applying TCL mothed on bornyl acetate of Amomum villosum Lour. medicine. Using GC method determine bornyl acetate contents in the essential oil of Amomum villosum Lour.; Using IR methed, GC-MS mothed to analyze infrared spectrum and essential oil GC-MS chromatographic analysis of Amomum villosum Lour.. Productspecification quantitative:On the basis of parameter determination in the character, using computer technology to study preliminary on division method of Amomum villosum Lour. products specification.3 ResultsChemical quality evaluation:3.1 31 batches of Amomum villosum Lour.'s water contents were all less than 15%, which matched the regulations of Chinese Pharmacopoeia. Determined 29 batches of Amomum villosum Lour.'s essential oil contents in the seed:samples collected in 2007-2008 basically matched the regulations of Chinese Pharmacopoeia, calculated on dry ingredients,most of them were more than 3%, majority of samples collected in 2009 were less than 3%. Applying TCL mothed on bornyl acetate of 19 batches Amomum villosum Lour. medicine.the samples of essential oil chromatography displayed color spots which were the same in corresponding chromatographic position of reference substance.3.2 27 batches of Amomum villosum Lour.'s bornyl acetate contents in essential oil were in the range of 0.4-0.6g/mL. Established the determination method of bornyl acetate in the essential oil of Amomum villosum Lour, preliminary.3.3 Amomum villosum Lour. from genuine producing regions and Yunnan,Guangxi, products of Amomum villosum had the same infrared absorption peak,they had no significant difference in the IR fingerprint. Analyzed 28 batches of Amomum villosum Lour.'s essential oil GC-MS chromatography, established the essential oil of Amomum villosum Lour. GC-MS chromatographic conditions and analysis methods,and identified ten characteristic components in essential oil.Product specification quantitativeUsing statistical methods to divide 100 fruits of Amomum villosum Lour. into three grads.4 Conclusion4.1 According to the experiment results,refering to the approach of 2005 version of《The People's Republic of China Pharmacopoeia》, initially established the chemical quality evaluation standards of Amomum villosum Lour. from genuine producing regions:the contents of water should less than 15.0%, calculated on dry ingredients,the contents of essential oil in the seed group should more than 3.0%,the contents of bornyl acetate (C12H20O2) in the essential oil of Amomum villosum Lour. should not less than 0.46g/mL, established TLC method of bornyl acetate of Amomum villosum Lour. and GC-MS chromatographic analysis methods of Amomum villosum Lour. essential oil.4.2 Preliminary establishing quantitative method of Amomum villosum Lour. product specification from genuine producing regions, the results have some reference value, new ideas for identification of Amomum villosum Lour. products are supposed.
