ObjectiveIn this study,we investigated the effects of Toll-like receptors and RIG-I-like receptor signaling pathway on the pathogenesis of innate immunity,we research the approach of innate immunity in HBV Transgenic mouse as well as a possible target and mechanism of The Nonrishing Kidney and Elearing Toxicity resisting HBV,to explore the scientific basis for the chronic hepatitis B and "Kidney deficiency and Evil lurk",and provide new ideas for the treatment of chronic hepatitis B by The Nonrishing Kidney and Elearing Toxicity.MethodsFifty HBV transgenic mice were randomly divided into five groups:a high-dose nourishing kidney and eliminating toxicity decoction group,a middle-dose nourishing kidney and eliminating toxicity decoction group and a low-dose nourishing kidney and eliminating toxicity decoction group,a modern medicine group,a HBV transgenic mouse model control group,and set up a ten normal mice as control group.The liver was collected from each mice,which kept a SPF room at the day of 60d,then the total RNA was extracted.The RNA was reversely transcribed into cDNA,and then perform real-time quantitative PCR in ABI 7500 Real Time PCR System to relative quantify the gene expression according to an internal control for GAPDH.The difference of Toll-like receptors and RIG-I-like receptor signaling pathway related molecules gene expression was compared between in each group by 2-??CT methods.Results1.The expression of Toll-like receptors and RIG-I-like receptor signaling pathway signaling pathway in HBV transgenic mouse model control group and normal mice:Compared with normal mice,the expression level of TLR-2?TLR-3?TLR-4?TLR-8 and TLR-9 mRNA in HBV transgenic mouse model control group was not significantly changed,without significant difference(P>0.05).Compared with normal mice,the expression level of RIG-I mRNA was increased and MDA-5 mRNA was decreased in HBV transgenic mouse model control group,and the difference was statistically significant(P<0.05),but the expression level of IPS-1 mRNA was not significantly changed,without significant difference(P>0.05).Compared with normal mice,the expression level of INF-??INF-??TNF-??TGF-??IL-6 mRNA in HBV transgenic mouse model control group was not significantly changed,without significant difference(P>0.05).but the expression level of MCP-1?IL-1? mRNA was decreased,and the difference was statistically significant(P<0.05).2.The expression of Toll-like receptors and RIG-I-like receptor signaling pathway related molecules gene expression in Nourishing Kidney and Eliminating Toxicit decoctiony influence HBV transgenic mouse:Compared with HBV transgenic mouse model control group,the expression level of TLR-2?TLR-3?TLR-4?TLR-8 and TLR-9 mRNA in the drug group was not significantly changed,without significant difference(P>0.05).And the expression level of TLR-8 mRNA in the middle-dose Nonrishing Kidney and Elearing Toxicity group was decreased,and the difference was statistically significant(P<0.05).Compared with HBV transgenic mouse model control group,the expression level of RIG-I mRNA in the middle-dose,the high-dose Nonrishing Kidney and Elearing Toxicity group was decreased,and the difference was statistically significant(P<0.05),and With the dose of Nonrishing Kidney and Elearing Toxicity group increase,RIG-I mRNA decreased.Compared with HBV transgenic mouse model control group,the expression level of MDA5 mRNA in the low dose,the middle dose and the high dose Nonrishing Kidney and Elearing Toxicity group was not significantly changed,without significant difference(P>0.05).Compared with HBV transgenic mouse model control group,the expression level of MDA5 mRNA in modern medicine group were increased,and the difference was statistically significant(P<0.05).Compared with HBV transgenic mouse model control group,the expression level of IL-1??MCP-1 mRNA in the low dose,the middle-dose and the high dose Nonrishing Kidney and Elearing Toxicity group was reduced,and the difference was statistically significant(P<0.05).Conclusions1.Liver damage in HBV transgenic mice was not associated with Toll-like receptors signaling pathway;and Nourishing Kidney and Eliminating Toxicit decoctiony did not affect the expression of Toll-like protein receptor.2.Nourishing Kidney and Eliminating Toxicit decoctiony can reduce the expression of IL-1? and MCP-1 mRNA in HBV transgenic mice,which may be one of the targets for the treatment of hepatitis B.3.RIG-I produce IL-1??MCP-1 may through the inflammasome signaling pathway,leading HBV transgenic mice liver damage.The mechanism of Nonrishing Kidney and Elearing Toxicity decoctiony regulating the innate immune function may be through regulating this signaling pathway and reducing the inflammatory response.4.The Nonrishing Kidney and Elearing Toxicity that Based on " Kidney deficiency and Evil lurk " pathogenesis treating HBV is not killing virus,but conducting long-term treatment by improving the body' s immunologic functions and reduce inflammation,to achieve the purpose of preserving the liver.This study offered scientific reference and new ideas for treating HBV with The Nonrishing Kidney and Elearing Toxicity and has instructional significance for clinic.
