Objective:To explore the effect of different doses of Kaixin powder on the learning and memory function and the expression of Bcl-2?Bax?Caspase-3 and P-P38MAPK/P38MAPK in the hippocampus tissue of Alzheimer' s rats mediated by A?-42.Investigate the mechanism and best dose of the anti-apoptotic action of Kaixin powder.Methods:1.Sixty clean level Wistar rats were randomized into five groups including blank group,model group,KP low-dose group,KP mid-dose group and KP high-dose group.Establish models with Alzheimer' s disease by injecting A? 1-42oligomer into bilateral hippocampus.After successfully modeling,the KP groups were given different doses of kaixin powder by gavage for four weeks.The other two groups were given the same volume distilled water.2.To observe the behavior change of the rats through Morris water maze test the day after the last treatment.3.To observe the neuronal apoptosis in the hippocampus tissue of Alzheimer' s rats by Tunel assay after the last day of Morris water maze test.4.To observe the expression of Bcl-2.Bax and Caspase-3 in the hippocampus tissue of Alzheimer' s rats by immunohistochemical method.5.To observe the expression of P-P38MAPK/P38MAPK in the hippocampus tissue of Alzheimer' s rats by western blot.Results:1.From the second day of orientation navigation experiments,the escaping latency was significantly shorter in the KP groups.The downward trend was more obvious in the high-dose group,with significant differences compared with the low-dose group and mid-dose group(P<0.05).In the observation of percentage of the second quadrant time and traversing times,it showed marked increasing in the KP groups.But a higher trend was showed in the high-dose group compared with the low-dose group and mid-dose group in percentage of the second quadrant time(P<0.05).While,there was no statistical difference in there KP groups in the observation of traversing times(P>0.05).2.The number of apoptosis cells in the KP group was obviously fewer than that in the model group.3.The expression of Bcl-2,compared with the model group,was increased in the KP group(P<0.01),and the high-dose group showed a better effect(P<0.05).The expression of Bax and Caspase-3,compared with the model group,was decreased in the KP group(P<0.05/P<0.01),and the high-dose group showed a better effect(P<0.05).4.The expression of P-P38MAPK/P38MAPK,compared with the model group,was decreased in the KP group,and the high-dose group showed a better effect(P<0.05).But there was no obvious difference between the low-dose group and the model group(P>0.05).Conclusions:1.The kaixin powder has a marked effect on ameliorating learning and memory ability of Alzheimer' s rats.2.The kaixin powder can improve learning and memory function of Alzheimer' s disease rats through up-regulating the Bcl-2 and down-regulating the Bax?Caspase-3 and p-p38/p38.The high-dose Kaixin powder showed a better effect.
