Object: To observe the changes of Bcl-2 and Bax expression, learning and memory abilities, TUNEL' S apoptosis cells in Alzheimer' s disease rats injected amyloid-beta protein at hippocampus and to investigate the protection and the mechanism of PMT on apoptosis of hippocampal neurons. Method: The AD rat model was established by infection of amyloid-beta protein into right hippocampus in rats. Learning and memory ability was studied by Y-eleetric maze, the expression of apoptosis related protein Bax and Bcl-2 in rats hippocampus were studied using immunohistochemistry method. The apoptosis neurons were studied by TUNEL' s method. Results: The model rats injured the right hippocampus by Ap, showed their disorders of learning and memory abilities. Bax is over- expressed and Bcl-2 is lower expressed, and typical apoptosis neurons in the CAi of hippocampus. The PMT group can improve the learning and memory abilities, the expression of Bax can be decreased and that of Bcl-2 can be increased significantly. The disposits of A P can only be seened in two rats of the model Conclusions: the aforesaid characteristic changes are similar to AD patients' some clinical and pathologic features, so that may be a valuable AD animal model. PMT was found to inhibit the apoptosis occurring in the model rats hippocampus and to improve disorders of learning and memory obviously.PMT might be beneficial for the prevention and therapy of Alzheimer's Disease.The neurotoxicity of A P might be not only through self-derect celluiotoxic effects, but also might through other mechanisms.
