Association Of Klotho Protein Serum Expression Level With NO And Polymorphism Of Klotho Gene In Ischemic Stroke Patients

Methods: 262 subjects were studied and divided into 2 groups,control group with 104 healthy people and ischemic stroke group(IS group)with 158 patients.In 262 subjects,all clinical data were collected.The genotyping of polymorphisms G-395 A,F352V,C370 S and C1818 T of the klotho gene was performed by allele-specific polymerase chain reaction assay(ASP-PCR).Klotho and NO level were measured by ELISA.Results: 1.In terms of age,gender,coronary disease,TG,TC,LDL,F352 V and C370 S,there was no difference between the two groups(p>0.05).Compared with control group,subjects of IS group had a significant higher rate associated with hypertension,diabetes,FG,HCY,G-395 A,C1818T,smoking and alcohol consumption(p<0.05).Subjects of IS group had a significant lower level in HDL,klotho and NO(p<0.05).2.Relative risk rate of subjects with hypertension to develop ischemic stroke increased by 1.8 folds compared with subjects without hypertension.Relative risk rate of subjects with smoking increased by 2.4 folds compared with subjects without smoking.Relative risk rate of subjects with alcohol consumption increased by 2.6 folds compared with subjects without alcohol consumption.Relative risk rate to develop ischemic stroke increased by 3.5% for HCY increased each unit,while decreased by 0.7% for NO increased each unit.3.There exit positive correlative relationship between klotho and NOin ischemic stroke patients.Conclusions: 1.G-395 A and C1818 T polymorphism of Klotho gene is likely to relate to the development of ischemic stroke.2.There is no correlative relationship between F352 V,C370S polymorphism of Klotho gene and the development of ischemic stroke.3.NO is a protect factor associated with ischemic stroke.4.klotho has a positive correlative relationship to NO in ischemic stroke patients.5.Hypertension,diabetes,smoking,alcohol consumption and hyperhomocysteinedemia are risk factors associated with ischemic stroke.
