Background and objective:Stroke is a kind of polygenic disease caused by genetic and environmental factors.Searching for predisposing genes of stroke is the focal point of study for genetic mechanism of stroke. KLOTHO gene is significantly associated with ageing-like phenotypes, atherosclerosis and so on,which are risk factors of stroke.So,KLOTHO gene may be the predisposing gene of stroke.Therefore,we studied the relationship between the SNPs of KL-VS Functional Variant of KLOTHO gene,F352V and C370S,and stroke for searched the genetic markers of stroke.Methods:The F352V and C370S of KLOTHO gene of 273 patients with cerebral infarction,205 patients with cerebral hemorrhage and 213 healthy individuals were genotyped with ASP-PCR.Meanwhile we tested the levels of blood pressure,serum lipid,blood glucose and BMI.All data was analyzed by SPSS 13.0.Results:1.The polymorphisms of the F352V and C370S of KLOTHO gene exist in Chinese people in Hunan province,and they are in complete linkage disequilibrium.2.The polymorphism of the F352V in CI group between with control group has significant difference(P<0.05),the percent of FF homozygous in CI group is significant higher than control group.In subgroup analyses of CI group,there is significant difference in F352V polymorphism between CI within hypertension with CI without hypertension,and similarly it is also found between CI with artery atherosclerosis plaque with CI without artery atherosclerosis plaque.3.No significant difference is found between CH group with control group in polymorphism of the F352V.(P>0.05).The results of CH group's F352V polymorphism subgroup analyses demonstrate that CH with CAD or DM is not significant different with CH without CAD or DM.4.The levels of blood pressure,blood glucose and blood lipids are significant different in control group with CI group and CH group.And the VV genotype subgroup has significant higher level of SBP and DBP than FF genotype or FV genotype.No difference in levels of TC,TG, LDL-C,HDL-C and FBS is found among genotype subgroups in statistics.Conclusions:1.The polymorphisms of the F352V and C370S of KLOTHO gene exist in Chinese people in Hunan province,and they are in complete linkage disequilibrium.2.The polymorphisms of KLOTHO gene F352V might be associated with cerebral infraction in Chinese lived Hunan province,no association with cerebral hemorrhage.3.The polymorphism of KL-VS variant might a risk of hypertension, cardiovascular disease and carotid atherosclerosis plaque for CI patients.
