Objective:The study is to investigate the possible relationship between two polymorphisms of SG13S114 (rs1050739) and SG13S32 (9551963) in the ALOX5AP and ischemic stroke.Methods:A case-control study was taken from 440 IS patients (the case group) and 486 non-IS controls (the control group)whch were matched for age and sex. The polymorphisms of SG13S114T/A and SG13S32C/A were analyzed by using polymerase chain reaction and Restrictive fragment length polymorphism(PCR-RFLP). The relationship of genetype and allele was analyzed by Statistic Package for Social Science. The association of the genotype and frequencies with the risk of ischemic stroke was analyzed by logistic regression to adjust for confounding variables.Result:(1)There were significant differences between the case group and the control group in SG13S114 genetype (X2=15.76, P<0.001),A allele was higher in the case group than in the control group,and the differences between them had statistics significance(X2=15.68, P<0.001).People with AA genetype have twice as high morbidity than people with non-AA genetype after the analyze of logistic regression to adjust for confounding variables (OR=2.035, P=0.006).In the following subgroup analyze,only in the LAA subgroup,AA genetype and A allele frequency are higher than the controls.The differences still has statistics significance after the analyze of logistic regression to adjust for confounding variables (OR=2.006, P=0.012).There was no difference of genetype and allele frequency between the SVD、CE subgroups and the controls.(2)There was no significant difference in the distribution of SG13S32C/A genetype and allele frequencies in the ischemic stoke and controls. In the following subgroup analyze,there was no difference of genetype and allele frequency between LAA、 SVD、CE subgroup and the controls.Conclusion:The polymorphism of SG13S114 in ALOX5AP gene may be involved in the risk of developing ischemic stroke.and the risk is from the A allele.There was no evidence that the polymorphism of SG13S32 is related to the ischemic stroke.
