Objective:This study is designed to investigate G395 A and F352 V polymorphism of Klotho gene in ischemic stroke patients sampled in the population of Han nationality in Jilin Province. By comparing the genotyping and allele frequencies of G395 A and F352 V, possible correlative relationship is supposed to be revealed between Klotho gene polymorphism and the development of ischemic stroke in northeast region of China.Methods:In Jilin Province, 415 subjects of Han nationality were studied and divided into 2 groups, control group and ischemic stroke group(IS group). There were 208 inpatients from Department of Neurology, the Second Hospital of Jilin University in IS group and 207 healthy people from physical examination center in control group. In 415 subjects, blood pressure, fasting plasma glucose, lipid profiles, carotid artery antherosclerosis index were measured. The genotyping of polymorphisms G395 A and F352 V of the Klotho gene was performed by allele-specific polymerase chain reaction assay(ASP-PCR). All data were processed by Excel 2007 and analyzed by SPSS 18.0.Results:1. Compared with control group, subjects of IS group had a significant higher rate associated with atherosclerosis, hypertension, diabetes, coronary disease, hyperlipidemia, hyperhomocysteinedemia, and smoking(p<0.05). In terms of gender and alcohol consumption, there was no difference between the two groups(p>0.05). Subjects of IS group had a significant higher level in FSB, TG, TC, Hcy, SBP, DBP(p<0.05). There was no difference in HDL-C and LDL-C level between two groups(p>0.05).2. In terms of genotyping of G395 A polymorphisms, there was no statistic difference between two groups(χ2=1.925, p=0.381). There was no statistic difference between two groups in terms of either G allele or A allele frequencies of G395 A polymorphism(χ2=2.065,p=0.151).In terms of GG genotype and GA+AA genotype distribution, there were no statistic differences between every two IS subgroups categorized by the risk factors(p >0.05). In terms of either G allele or A allele frequencies of G395 A polymorphism, there was no statistic difference between every two IS subgroups categorized by the risk factors(p>0.05).3. There was a significant difference between two groups in terms of genotyping of F352 V polymorphisms(p<0.05). Compared with control group, both FF genotype and F allele frequencies of F352 V polymorphisms were significantly higher in the IS group(χ2=9.519, p=0.009 & χ2=3.907, p=0.048 respectively).In IS subgroups categorized by the risk factors, both FF genotype and F allele frequencies of F352 V polymorphisms were significantly higher in hypertension and atherosclerosis subgroups(p<0.05 & p<0.05 respectively).4. There were no statistic differences for mean value of FSB, TG, TC, HDL-C, LDL-C, Hcy, SBP and DBP between every two subgroups of GG genotype, GA genotype and AA genotype of G395 A polymorphism either in IS group and control group(p>0.05).There were no statistic differences for mean value of FSB, TG, TC, HDL-C, LDL-C, Hcy,SBP and DBP between every two subgroups of FF genotype, FV genotype and VV genotype of F352 V polymorphism either in IS group and control group(p>0.05). Mean value of SBP is significantly higher in VV genotype of F352 V polymorphism subgroup compared with both FF genotype and FV genotype subgroup(p<0.05).5. According to logistic regression analysis, risk factors associated with ischemic stroke are atherosclerosis, hypertension, and hyperhomocysteinedemia. Among them, relative risk rate of subjects with hyperlipidemia to develop ischemic stroke increased by 2.9 folds compared with subjects without hyperlipidemia. Relative risk rate of subjects with hypertension increased by 1.2 to 4.9 folds compared with subjects without hypertension, and relative risk rates increased with the severity of hypertension. Relative risk rate of subjects with atherosclerosis to develop ischemic stroke increased by 7.2 to 17.5 folds compared with subjects without atherosclerosis, and relative risk rates increase with the severity of atherosclerosis. Relative risk rate of subjects with hyperhomocysteinedemia to develop ischemic stroke increased by 1.6 folds compared with subjects without hyperhomocysteinedemia.6. According to logistic regression analysis, in terms of relative risk rates to develop ischemic stroke, there were no statistic differences between every two subgroups of GG genotype, GA genotype and AA genotype of G395 A polymorphism either in IS group and control group(p>0.05).7. According to logistic regression analysis, there were significant statistic differences between every two subgroups of FF genotype, FV genotype and VV genotype of F352 V polymorphism either in IS group and control group(p<0.05). Relative risk rates to develop ischemic stroke was higher in FF genotype of F352 V carriers compared with either FV genotype carriers or non-carriers. Relative risk rates to develop ischemic stroke in FF genotype of F352 V carriers increased by 1 time compare with either FV genotype carriers or non-carriers.Conclusions:1. There is no correlative relationship between G395 A polymorphism of Klotho gene and the development of ischemic stroke in the Han nationality population in Jilin province.2. F352 V polymorphism of Klotho gene is related to the development of ischemic stroke in the Han nationality population in Jilin province. FF genotype carriers has a higher risk to develop ischemic stroke than non-carriers.3. F352 V polymorphism of Klotho gene may be related to the development of hypertension in ischemic stroke patients, especially systolic blood pressure elevation in Han nationality population in Jilin province.4. F352 V polymorphism of Klotho gene may be related to atherosclerosis in ischemic stroke patients in Han nationality population in Jilin province.5. Risk factors associated with ischemic stroke are atherosclerosis, hypertension, and hyperhomocysteinedemia in Han nationality population in Jilin province.6. Allele polymorphism in F352 V of Klotho gene may be a dependent risk factor of ischemic stroke in Han nationality population in Jilin province. |
