Background:Type 2 diabetes mellitus is a kind of metabolic diseases characterized by chronic hyperglycemia caused by multiple causes.It is not only have a high incidence,and the prevalence rate of it also showed an upward trend.Complications of it are diverse,and cardiovascular and cerebrovascular disease is one of its major complications.The incidence of atherosclerosis in the blood vessels of type 2 diabetes is significantly higher than that in patients without type 2 diabetes,which is one of the important causes of death.Early regulation of blood glucose,blood lipids and prevention or mitigation of arterial disease is an important method for the treatment of type 2 diabetes and blood vessels diseases.Objective:To observe the effect of Modified Si-Miao-San on blood glucose,blood lipid and aortic lesions in animals' T2DM model for the clinical application.Methods:The experiment is divided into two parts.Experiment one:After one week of accommodation in mice with high lipid diet plus streptozotocin(STZ)to replicated the model of type 2 diabetic mice.After one week,the mice were trailing blood.Fast blood glucose(FBG)with a blood glucose meter was tested,and then the mice with FBG?11.1mmol·L-1 were selected.The mice which were modeled successfully will be divided into five groups:model group and control group given distilled water,Modified Si-Miao-San high,medium and low dose group,metformin group fed to the corresponding drugs,once a day for three weeks.Glucose and serum TC,TG,HDL-C,LDL-C content were tested.And the hypoglycemic and lipid-lowering effect of Modified Si-Miao-San for type II diabetic mice was observed.At the same time the liver tissue pathological examination of liver was taken.Experiment two:60 rats were divided into 6 groups randomly:Positive group,model group,blank group,low?middle and high dose SiMiao group.Except the blank group,other groups fed with mash(18%lard,7%cholesterol,1%bovine bile salt,74%peanut oil)once a day.After eight weeks of modeling filling the mash was continued.After six weeks,the rats were fasting 12h,and then blood content of CHOL,TG,Glucose LDL-C?HDL-C,CGRP,Rsistin were tested.Results:The first part of the experimental results show that compared with the control group,blood glucose and lipid levels of model group were significantly elevated(P<0.01).The blood sugar levels of the model group were significantly decreased compared with Modified Si-Miao-San low,medium and high dose groups.(P<0.05,P<0.01),and the lipid levels in Modified Si-Miao-San middle and high dose groups were significantly decreased(P<0.01),while in low dose group TG,HDL-C,LDL-C levels were no significant differences.The second part of the experimental results show that Compared with the blank control group,the levels of serum CHOL,TG,Glucose in the model group were elevated(P<0.05,P<0.01),and LDL level was elevated(P<0.01)At the same time,pathology results showed that,Hyperlipidemia rats' aortic atherosclerosis occurs early lesions.This proves that the experiment successfully replicated the early models of atherosclerosis.And the positive group,Simiaokeli group(low,medium and high level)showed good reduction in the content of LDL-C,resistin andincreasing of HDL-C and CGRP's Pathology results also show improvement in their aorta situationConclusion:The Modified Si-Miao-San can reduce the levels of blood glucose and blood lipid in model mice of type 2 diabetes mellitus,and can reduce the level of resistin in arteriosclerosis rats and increase the level of calcitonin gene-related peptide(CGRP),and interfere with arteriosclerosis of diabetic macrovascular.
