Objective: To create the rabbit knee joint traumatic synovitis in animal models,application of Si Miao San to intervent whose levels of interleukin1β (interleukine1β,IL-1β) and tumor necrosis factor alpha (tumor necrosis factor α and TNFα),and to observethe morphological changes. Aims to:①explore the efficacy mechanisms of Si Miao San onthe rabbit knee traumatic synovitis from the molecular and cellular level.②provide a goodexperimental basis and theoretical basis for traditional Chinese medicine treatment oftraumatic synovitis of the knee Joint (Traumatic Synovitis of Knee Joint, TSKJ).Methods:24rabbits were randomly divided into three groups, Control group8,modelgroup8and the test group8. Started from the first day, the Control group and modelgroup were treated with saline gavage, and the test group was treated with Si MiaoSan(liquid concentration for1g/ml), gavage dose4.7ml/kg,2times/d, a total of10d. To11days observed joint circumference changes, joint surface temperature changes, and thenanimals were sacrificed to extract the synovial fluid, using enzyme-linked immunosorbentassay (enzyme-linked immunosorbent assay, ELISA) to detect the rabbit knee joint fluidIL-1β and TNF-α levels. Patella line the outer edge of the longitudinal incision, take thesynovial tissue from the junction of the patella and femur, the specimens were fixed in10%formalin solution, the48h experts paraffin sections, and observe the synovial tissuebiopsy to explore inflammation the expression levels of cytokines in knee joint synovitis inthe pathological process and its cytological changes.Results: The joint circumference of Control group compared with model group andtest group was significant (P<0.05); There were no significant difference between modelgroup and test group (P>0.05); The temperature of the articular surface of Control groupcompared with model group and test group was significant (P<0.05); There were nosignificant difference between model group and test group (P>0.05); For the IL-1β andTNF-α levels of inflammatory factors of the rabbit knee joint synovial fluid,there weresignificant difference between the control group and model group (P<0.05); There were also significant difference between model group and test group (P<0.05);The experimentshows that there was almost no inflammatory response in the Control group, there wereinflammatory response in model group and test group.According to the degree ofinflammation of the three groups under the microscope, combined with the rabbit kneejoint biopsy results showed that Control group compared with model group and test groupwas significant (P<0.05); there were significant difference between the control group andmodel group (P<0.05);Conclusion:1. Si Miao San have a better therapeutic effect on the rabbit knee jointtraumatic synovitis.2. Si Miao San can inhibit IL-1β and TNF-α levels, which play the roleof the treatment of traumatic synovitis.
