Intervention Study On The Method Of Yiqi Yangyin Huayu Tongluo In Thoracic Aorta Disease Of T2DM Rats

This thesis is chiefly departed into two parts of theoretical and experimental study. The literature theory study is a systematic study of traditional Chinese medicine and Western Medicine on type 2 diabetes mellitus(T2DM) macrovascular lesions and also a in-deepth theoretical discussion of Yiqi Yangyin Huayu Tongluo law on the prevention and treatment of T2DM macrovascular lesions. Part of the experiment, T2DM rat model was established to observe the effect of Tongxinluo on thoracic aorta in T2DM rats.Objective:To study the effect of the function of Tongxinluo on T2DM rats fasting plasma glucose (FPG), blood lipid, superoxide dismutase (SOD),malondialdehyde (MDA) and the reactive oxygen species (ROS), advanced glycosylation end products (AGEs),protein expression in rat thoracic aorta, to explore the mechanism of Yiqi Yangyin Huayu Tongluo method for preventing and treating T2DM macrovascular disease.Methods:80 male Wistar rats were fed accommodately for 1 week,and then stochasticly departed into experimental group (70 rats) and control group (10 rats).The experimental group fed with high-fat diet, control group with conventional standard diet, for 4 weeks,a total of 5weeks. At the end of fifth week, by intraperitoneal injection of fresh configuration (30mg/kg 1%STZ) citric acid sodium citrate buffer in the experimental group rats,and the same dose of citric acid sodium citrate buffer in the blank control group.3 days later,measuring the fasting blood glucose of rats tail blood,two successive measuring more than 16.7mmol/L, as the success of modeling.Remove the rat model of failure and death, the successed model of Wistar rats remained to 56,which were randomly divided into 4 groups:model group (n= 14), simvastatin group (n= 14), Tongxinluo high-dose treatment group (n= 14), Tongxinluo low dose treatment group (n= 14),continued feeding with high sugar and high fat diet untill ninth week. Then, high and low dose Tongxinluo group and simvastatin group rats were countinued administering till twenty-first week. After that.detected the FPG, blood lipids, SOD, MDA, ROS, AGEs levels and the expression of AGEs in thoracic aorta were detected at the end of twenty-first week.Results:In the respect of weight, high-dose of Tongxinluo treatment group is higher than the model group and the simvastatin group (P<0.01); FPG, before treatment, model group and treatment group and control group compared with the normal group were observably increased (P< 0.01); after treatment, compared with model group,control group and treatment group were not observably decreased (P>0.05); Blood lipid, model group TG, TC and LDL-C compared with the normal group were obviously increased (P<0.05), the content of HDL-C was obviously decreased (P<0.05), compared with the model group, the rats in the treatment group TG, TC, LDL-C all decreased (P<0.05), HDL-C increased observably (P<0.05), high-dose Tongxinluo treatment group rats’ serum TG observably reduced (P<0.05),compared with the control group,also HDL-C increased observably (P<0.05), LDL-C decreased observably (P<0.05); SOD activity, model group was observably lower than normal group (P<0.01), compared with the model group, the treatment group increased observably (P<0.05), high-dose Tongxinluo group was higher than the simvastatin group(P<0.05); MDA, the model group was observably higher than the normal group (P<0.05), the treatment group compared with the model group was observably lower (P<0.05), high-dose Tongxinluo group was lower than the simvastatin group (P<0.05); ROS and AGEs, the treatment group compared with the model group raised observably (P<0.05); the expression of AGEs, compared with the model group, the treatment group was observably dropped (P<0.01), high and low dose of Tongxinluo treatment group compared with simvastatin group were observably decreased (P<0.05).Conclusion:Tongxinluo can observably improve the level of atherosclerosis, so as to effectively treat or alleviate T2DM macrovascular lesions.The possible mechanism of Tongxinluo:increased SOD activity, reduced the accumulation of ROS, AGEs and MDA in the blood vessels, down-regulated AGEs protein expression and so on. This method can regulate rats’ blood lipid metabolism, enhance the antioxidant capacity and improve the metabolic memory effect in high glucose rats to reduce macrovascular disease.This study not only can provide important theoretical guidance for Yiqi Yangyin Huayu Tongluo therapy in the treatment of T2DM macroangiopathy, but also provides a good foundation for the future use of Chinese medicine in the prevention and treatment of T2DM thoracic aortic pathologies.
