ObjectiveUsing the metabolomic liquid mass spectrometry to healthy people,liver-kidney yin deficiency type and spleen kidney yang asthenia type of urine in patients with systemic lupus erythematosus(sle)full spectrum analysis,analysis the differences metabolites of two type of sle,to find out the corresponding potential biomarkers and trace the metabolic pathways of important biomarkers,discuss two type objective differences and TCM syndrome type connotation,for the two card type to the nature of objective and system research provided the scientific basis.Methods60 SLE patients,who visited the doctor at the outpatient clinic of Guangdong Province Traditional Medical Hospital dermatological department from December of 2015 to January of 2016,have been encompassed into research.Spleen kidney yang asthenia type and liver-kidney yin deficiency type.30 each;30 patients of the healthy control group were all from the physical examination center.Collecting morning midstream urine and having centrifugal treatment,then transferring and numbering 1.5 mL of this solution,cryopreserving it.Before experiments,after thawing and refreezing it at 4?,preparing the QC samples and other various samples.The Data collection and sample treatment were all based on the software,Waters UPLC-QTOF-MS.The DataRow pretreatment was based on the MakerLynx XS and then input the peak data into the recognition processing software,SIMCA-P+ 11.5.Through pattern recognition to complete the PCA and PLS-DA analysis and screening potential biomarkers through VIP value and identifying them through previous literature research and chemical measurement.ResultesThrough the mass spectrometry outline,spleen kidney yang asthenia type and liver-kidney yin deficiency type can be clearly distinguished from the control group.These two have the same.There are also different.Through the correlation appraisal to determine the potential biomarkers.which are Glycine,Phenylpyruvic acid,N-Formylkynurenine,Octadecanol,Alpha-Hydroxyhippuric,Pantothenic acid,triethyl citrate,20-hydroxy-LTE4,N-Acephate,Petroselinic acid,S-(Hydroxymethyl)glutathione,a-ketoglutarate.ConclusionsThere is a significant difference in endogenous small molecule metabolites between the healthy and the SLE patients which can be presented obviously under the techniques of UPLC-MS.SLE patients have two different types which were show up in potential biomarkers.Patients of spleen kidney yang asthenia type and liver-kidney yin deficiency type have obvious different metabolites which include serine metabolism,tryptophan metabolism,phenylpyruvic acid metabolism,intestinal flora metabolism,tricarboxylic acid cycle and fatty acids metabolism,etc.
