Objective:"Zeng Ye Rong Chang Fang" is Professor Li Feng prescription,clinical application has intestinal laxative effect and the treatment of functional constipation,also can be used as adjuvant to chronic renal failure enema,and have a certain therapeutic effect on melanosis coli.The purpose of this experiment is to verify the treatment effect: 1.increase of intestinal fluid wing constipation rats to demonstrate the efficacy of TongFuXiezhuo;2.to investigate the regulating effect of Zengye decoction on expression of AQP2 in colonic intestinal wing of rats,to study the mechanism of TongFu;3.Zeng Ye Rong Chang Fang modulatory effects on renal expression of intestinal side AQP2 rats and study its role in protecting kidney.Methods:60 healthy male SD rats,this experiment selects the weight 160-200 g,10rats were randomly selected as normal group,the rest were model group,model group SD rats were given compound diphenoxylate suspension gavage model,continuous intragastric administration of 8 weeks,given to each rat by gavage in 5% ink indicator after intragastric administration of 30 min each rat,and recorded the first time of defecation,stool weight and grain number 12 h,statistical analysis was performed on rats defecation time,grain number and weight of stool 12 h was statistically significant compared with normal group until the model is.Then the model rats were randomly divided into 5 groups,namely,model group,control group,add liquid Rong Fang Fang low,high,medium dose group,each group of 8 selected;the normal group was randomly selected for only 8.Were divided into 6 groups.The normal group was given distilled water 1ml/(100g?d)BW,1/d,gavage;model group were given normal saline1ml/(100g?d)BW,1/d,the control group was given intragastric administration;MaZiRenWan 0.536g/(kg?d)BW,1/d,intragastric liquid;Zeng Ye Rong Chang Fang low dose group,Zeng Ye Rong Chang Fang granule 2.24g/(kg?d)BW,1/d,i.g.;in middle dose group,Zeng Ye Rong Chang Fang granule 4.84g(kg?d)BW,1/d,i.g.;high dose group,Zeng Ye Rong Chang Fang granule 9.68g/(kg?d)BW,1/d,i.g..After 4 weeks of intragastric administration,the rats werecollected and weighed,and the time of the first black stool excretion was calculated.The pathological changes of colon and kidney of rats in each group were observed by light microscope,and the expression of AQP2 in intestine and kidney was detected by Western-blot and immunohistochemistry.Results:The results showed that the rats in model group appeared irritability,poor mental status,activity,or get together,loss of hair,dry stool,discharge difficult,hard water reduced phenomenon.The normal group was in good condition.After administration,the state of the rats in each administration group was improved,the symptoms were relieved,and the intake of water was increased.By observing the rats feces found,compared with the model group,control group,Zeng Ye Rong Chang Fang low dose group,middle dose group and high dose group significantly shorten defecation time,defecation quantity increases,ink pushing rate increased significantly.Through the light microscope observation of colon and kidney tissue of rats showed changes of normal rats colonic mucosal epithelial cells arranged regularly,mucosa ulcer,inflammation,muscle layer neat without thinning,other rats colonic mucosal epithelium were arranged irregularly,the high dose group compared with those of the normal group.In the model group,the epithelial cells were arranged irregularly,the muscle layer became thinner,the blood vessels expanded,and the inflammatory cells infiltrated.Compared with the model group,the mucosal epithelial cells in each administration group were gradually arranged in order,the vascular dilatation was reduced,the inflammatory cells were reduced,and the high dose group was higher than that in the normal group.The normal group of renal glomeruli were normal,no hyperplasia,normal tissue structure,glomerular structure clear,tubular structural integrity;model group glomerular erythrocyte aggregation,infiltration of inflammatory cells,glomerular cysts shrink or even disappear,glomerular basement membrane thickening,tubular lumen edema;the control group and the model of glomerular capillary expansion renal tubular edema is not obvious.In low dose group and model group,the inflammatory cell infiltration was obvious,and the glomerular capsule cavity was irregular.In the medium dose group and the model group,the glomerulus was normal,the size of the cyst cavity was normal,and the inflammatory infiltration was decreased.Compared with the model group,there was no obvious dilatation ofthe blood vessels in the high dose group,the size of the cyst cavity was the same,there was no inflammatory infiltration,and the renal tubular edema was not obvious.Detected by Western-blot and immunohistochemical methods visible,colonic AQP2 protein of rats in model group were significantly increased,and high dose group rat colon AQP2 protein and normal group,Zeng Ye Rong Chang Fang intestinal side decreased significantly;compared with the model group,the rats in the normal group,Zeng Ye Rong Chang Fang intestinal side high dose group kidney AQP2 obviously,while the control group,low dose group was decreased.Conclusion:1 The decoction can improve the general state of the rats with constipation,shorten the defecation time,increase the number and quantity of stool,and increase the amount of fluid to the intestinal bowel movement has a certain role in promoting.2 Zeng Ye Rong Chang Fang has a downward effect on the expression of AQP2 in rat colon,and has a certain effect on colonic pathology in rats with constipation.It further verified the effect of the decoction for promoting fluid retention and retention of intestine,which provides a new theoretical basis for the theory of " Tong Fu Xie Zhuo ".3 Zeng Ye Rong Chang Fang decoction has effects on intestinal wing up-regulated the expression of AQP2 in rat kidney,speculated that increasing liquid can play a role in protecting the intestinal wing of the kidney by regulating AQPs,which is to treat chronic kidney disease and related diseases by regulating the renal AQP2 provides new ideas and prescriptions.
