| Purpose:To explore the correlation pleural effusion removal by warming Yang to resolve fluid-retention therapy and expression of aquaporin 2 in kidney, to reveal its possible mechanism of water metabolism.Methods:40 healthy adult Wistar rats were randomly divided into four groups including the yang-warming and fluid retention-resolving therapy group (W), wiping off the cassia twig fluid retention-resolving group(Q), model control group(M) and bank control group (K), The pleural effusion mold was formulated by 1% Carrageenan injection and was evaluated by X-ray irradiation. The treatment was applied in two hours after molding for incessant 5 days by once a day.The blank control group and model group were not given any treatment, only the normal feed water supply. Then the rats were killed on the fifth day after molding.Their kidney tissue and plural effusion were collected. Using the western blot techniques for detecting the expression of AQP2, using enzme-linked immune sorbent assay(ELISA) determination of cAMP and PKA, and the results were compared between each group.Results:(l)Plueral effusion:Compared with the K group, the content of pleual effusion in other groups are all increased (P<0.05). Compared with the M group, the W group and Q group are all decreased after given the medicine (P<0.05). There is no significance,compared with the Q group, the content of pleual effusion in W group is increased (P>0.05).(2)AQP2:Compared with the K group, the expression of AQP2 in M group is decreased(P<0.05); Compared with the M group, the expression of AQP2 in W group and Q group are increased(P<0.05);There is no significance,though compared with the Q group, the expression of AQP2 in W group is increased (P>0.05).(3)PKA:Compared with the K group, the expression of PKA in W group and Q group are increased(P<0.05); There is no significance in the other groups (P>0.05).(4)PKCã€cAMP:Compared with the Q group, the expression of cAMP in W group are increased(P<0.05); There is no significance in the other groups (P>0.05).(5)Ultrastructure of Kidney:Compared with the K group, the ultrastructure of Kidney in M group have abnormal expression significantly;Compared with the M group, the ultrastructure of Kidney in W group and Q group abnormal expression relieve.Between W group and K group,there has no difference in the ultrastructure of Kidney.Conclusion:(1)The representative prescription of yang-warming and fluid retention-resolving therapy and the prescription of wiping off the cassia twig fluid retention-resolving have a positive influence in treating the model of pleural effusion about rat caused by λ-Carrageenan.(2) In the model of pleural effusion about rat caused by λ-Carrageenan, the expression of AQP2 in M group was reduced, The representative prescription of yang-warming and fluid retention-resolving therapy and the prescription of wiping off the cassia twig fluid retention-resolving can increase the expression with rats in pleural effusion models, it may be one of the mechanism to treat the disease of retained fluid.(3)The representative prescription of yang-warming and fluid retention-resolving therapy and the prescription of wiping off the cassia twig fluid retention-resolving can increase the expression of PKA in the model of pleural effusion about rat caused by X-Carrageenan,then adjustment AQP2,.Thus strengthen kidney on fluid reabsorption it may be one of the mechanism to treat the disease of retained fluid.(4)There is no significant influence on cAMP and PKA with the representative prescription of yang-warming and fluid retention-resolving therapy and the prescription of wiping off the cassia twig fluid retention-resolving.(5)The ultrastructure of Kidney in the model of pleural effusion about rat caused by λ-Carrageenan have abnormal expression significantly.The representative prescription of yang-warming and fluid retention-resolving therapy and the prescription of wiping off the cassia twig fluid retention-resolving have benign and promote recovery function on the abnormal expression.The representative prescription of yang-warming and fluid retention-resolving therapy have more significant effect. |
