Influence Of Yang-warming And Fluid Retention-resolving Therapy On Expression Of Aquaporin-4 And CAMP-PKA/PKC Pathway In Small Intestine Among Rats With Pleural Effusion

Pu r pose:To explore the influence of yang-warming and fluid retention-resolving therapy and wiping off the cassia twig fluid retention-resolving theapy on expression of aquaporin-4(AQP4)、cyclic adenosine monophosphate(cAMP), protein kinase A(PKA) and protein kinase C(PKC) in small intestine among rats with pleural effusion, to reveal its possible mechanism of water metabolism.Methods:Using the digitally random methods to divides the adult Wistar rats into four groups, the yang-warming and fluid retention-resolving therapy group (W), wiping off the cassia twig fluid retention-resolving group(Q), model control group(M) and blank control group (K), each group with ten rats(five male and five female per group). All rats expect those in blank control group received intrapleural injection of 1% carrageenan to establish a rat model of pleural effussion(pleural fluid retention). At 24 h after the model was established, model rats were subjected to pathological evaluation by radiography.At 2h after the model was established,each group received respective treatments by gavage once daily for 5 d. All rats were sacrificed after intragastrically administration, each side of the plural effusion and small intestine were collected. Using the western blot techniques for detecting the expression of AQP4, using enzme-linked immune sorbent assay(ELISA) determination of cAMP and PKA, and the results were compared between each group.Results:①Plueral effusion:Compared with the bank control group, the content of pleual effusion in other groups are all increased. Compared with the model control group, the yang-warming and fluid retention-resolving therapy group (W) and wiping off the cassia twig fluid retention-resolving group(Q) are all decreased after given the medicine. There is no significance between the W and Q group.②AQP4:Compared with the blank control group, the expression of AQP4 in model control is decreased; the yang-warming and fluid retention-resolving therapy group and wipe off the cassia twig fluid retention-resolving group is increased; there was no difference between the yang-warming and fluid retention-resolving therapy group, the wipe off the cassia twig fluid retention-resolving group and the blank control group.③cAMP:The content of cAMP in each group was no significant difference④PKA:Compared with the blank control group, the expression of PKA in other group are significance increased, and other group has no difference.⑤PKC:The content of PKC in each group was no significant difference.⑥The transmission electron microscope of small intestine indicate that rich microvilli on the surface of epithelial cells, mitochondrion swelling obviously, after the medicine was given, the microvilli on the surface of epithelial cells are closely packed, mitochondria, ribosome and rough endoplasmic reticulum are revealed clearly.Conclusion:(1)λ-carrageenan can manufacture pleural effusion sussceffully.(2) The yang-warming and fluid retention-resolving therapy group and wiping off the cassia twig fluid retention-resolving group have a positive influence in treating plural effusion.(3)In the model of pleural effusion about rat caused by λ-Carrageenan, the expression of AQP4 in empty intestine was reduced, the yang-warming and fluid retention-resolving therapy can increase the expression with rats in pleural effusion models, it may be one of the mechanism to treat the disease of retained fluid.(4)In this expreriment, the cAMP-PKA/PKC pathway may not the key to control the expression of AQP4 in small intestine.(5)The small intestine change the microvilli on intestinal epithelial cell particapte the yang-warming and fluid retention-resolving therapy to treat the pleural effusion.
