Objective:In this study,we used 10mg/ml areca nut extract(ANE)to induce oral submucous fibrosis in rats and treated with modified Jiawei Danxuankoukang to detect the expression of DKK3 and β-catenin in oral mucosa of OSF rats before and after treatment,and to explore the possible mechanism of modified Jiawei Danxuankoukang against oral submucous fibrosis.Methods:Fifteen SD rats were randomly divided into three groups:NC group was normal group,Model group was OSF model group,DXKK group was Jiawei Danxuankoukang group,every 5rats were a group.NC group did not do any treatment,but Model group,DXKK two groups of SD rats were used areca nut extract for modeling,modeling time for 8 weeks.After 8 weeks,rats in NC group and Model group were given distilled water for 4 weeks,and rats in DXKK group were given Jiawei Danxuankoukang for 4 weeks.After 4 weeks of administration,the rats were killed and buccal mucosa specimens were collected for later use.The expression of DKK 3 andβ-catenin protein was detected by immunohistochemistry and Western blot.The expression of DKK3 and β-catenin gene was detected by PCR.Results:(1)The results of immunohistochemistry showed that the positive expression of DKK3 and β-catenin in OSF model group was increased compared with normal group.Compared with OSF model group,the positive expression of DKK3 and β-catenin in Jiawei Danxuankoukang group decreased.(2)Western blot analysis showed that the expression of DKK3 and β-catenin protein in OSF model group increased compared with normal group.Compared with the OSF model group,the expression of DKK3 and β-catenin protein in Jiawei Danxuankoukang group were decreased,and the difference was statistically significant(P<0.05).(3)RT-q PCR results showed that:Compared with the normal group,the expression of DKK3 and β-catenin m RNA in OSF model group increased.Compared with the OSF model group,the expression of DKK3 andβ-catenin m RNA in Jiawei Danxuankoukang group were significantly decreased(P<0.05).Conclusion:(1)The expression of DKK3 in OSF was significantly increased,indicating that DKK3 may be involved in the formation of OSF.however,that increase expression of DKK3 and β-catenin in OSF suggest that DKK3 may have Wnt-independent function,and DKK3 may not act as a Wnt antagonist in the development of OSF.(2)Jiawei Danxuankoukang could improve the OSF induced by ANE,and decrease the expression of DKK3 and β-catenin,which may be one of the mechanisms of Jiawei Danxuankoukang in treating or improving the occurrence and development of OSF. |
